Floerkea proserpinacoides (Limnanthaceae) is a spring ephemeral annual species that grows in deciduous forests throughout eastern North America. Seeds germinate from late November to December, although the first leaf emerges only from late March to early April. Growth begins in early April at the onset of favourable temperatures, following snowmelt, and continues through mid-June. Senescence coincides with increasing air temperature and decreasing light level as a result of canopy closure. In this paper, we present the results of a growth chamber study designed to determine the effect of light level on growth, biomass allocation and reproduction of F. proserpinacoides. The study consists of two parts: in a first experiment, plants were grown at five constant photosynthetic photon fluence rates (PPFR: 90, 180, 360, 540 or 900 μmol m−2 s−1), and in a second experiment, PPFR was reduced from 900 μmol m−2 s−1 to 180 μmol m−2 s−1 after 0, 14, 21, 28 or 35 d of growth. Relative humidity, temperature, nutrient and water supply were kept constant in a hydroponic sand culture experiment. Total biomass, leaf mass and leaf area increased with increasing PPFR up to 540 μmol m−2 s−1. Plants grown at the highest (900 μmol m−2 s−1) and the lowest (90 μmol m−2 s−1) PPFR had a substantially lower biomass by the end of the 35-d growth period than plants grown at intermediate PPFRs (360 or 540 μmol m−2 s−1). Despite differences in total biomass, there were no significant differences in seed production among treatments. The mean relative growth rate (RGR) increased with increasing light levels between 90–540 μmol m−2 s−1, and it was reduced at 900 μmol m−2 s−1. However, differences in RGR were not significant among treatments. Specific leaf area did not vary consistently as a function of light level, whereas leaf area ratio and leaf mass ratio tended to increase with increasing PPFR, reaching maximum values at 360–540 μmol m−2 s−1. However, none of these growth variables differed significantly across the range of PPFR levels. The transfer of plants to lower PPFR had no significant effect on any of the growth components. Biomass production for the species appeared to be optimized at PPFR of 360–540 μmol m−2 s−1. Growth might be restricted by an insufficient supply of photosynthates at low PPFR and by photoinhibitory processes at higher PPFRs.

Northern red oak in the western Lake States area of the USA exists on the most xeric edge of its distribution range. Future climate-change scenarios for this area predict decreased water availability along with increased atmospheric CO2. We examined recent photosynthate distribution and growth in seedlings as a function of CO2 mole fraction (400, 530 and 700 μmol mol−1 CO2), water regime (well watered and water-stressed), and ontogenic stage. Water stress effects on growth were largely offset by elevated CO2.

Water stress increased root mass ratio without concurrently increasing allocation of recent photosynthate to the roots. However, apparent sink strength of water-stressed seedlings at the completion of the third growth stage tended to be greater than that of well watered seedlings, as shown by continued high export, which may contribute carbon reserves to support preferential root growth under water-stressed conditions.

Elevated CO2 decreased apparent shoot sink strength associated with the rapid expansion of the third flush. Carbon resources for the observed enhanced growth under elevated CO2 could be provided by enhanced photosynthetic rate over an increased leaf area (Anderson & Tomlinson, 1998, this volume).

Increased sink strength of LG seedlings under water-stressed conditions, together with decreased apparent shoot sink strength associated with growth in elevated CO2 provide mechanisms for offsetting water stress effects by growth in elevated CO2.

Careful control of ontogeny was necessary to discern these changes and provides further evidence of the need for such careful control in mechanistic studies.

Twelve-day-old soybean plants were exposed to atmospheric NO2 (0·3 μl l−1) and simultaneously supplied, via the roots, with 5 mM or 1 mM of NaNO3 or NH4Cl. After exposure for 7 d, the amount of NO2 absorbed per plant was greater in plants supplied with nitrate than in plants supplied with the same concentration of ammonium. The NO2 AR (absorption rate) decreased with increasing exposure time. At the beginning of exposure, the NO2 AR for all plants was c. 12 mg NO2 h−1 m−2 μl−1 l. On the day 7 of exposure, the NO2 AR declined to 8·46, 8·97, 8·27, and 9·04 mg NO2 h−1 m−2 μl−1 l for plants receiving 1 mM ammonium, 1 mM nitrate, 5 mM ammonium, and 5 mM nitrate respectively. The plants supplied with nitrate had a higher concentration of leaf nitrate and a higher pH than those supplied with the equivalent concentration of ammonium. These results suggest that the NO2 absorption rate might be attenuated by the accumulation of H+ produced from N uptake and assimilation.

Symploca PCC 8002 Kützing is a filamentous cyanobacterium that lacks the specialized cells, known as heterocysts, that protect nitrogenase from O2 in most aerobic N2-fixing cyanobacteria. Nevertheless, Symploca is able to carry out N2 fixation in the light under aerobic conditions. When cultures were grown under light/dark cycles, nitrogenase activity commenced and increased in the light phase and declined towards zero in the dark. Immunolocalization of dinitrogenase reductase in sectioned Symploca trichomes showed that the enzyme was present only in 9% of the cells. These cells lacked any obvious mechanical protection against atmospheric O2 and their ultrastructural characteristics were similar to those of cells that did not contain any dinitrogenase reductase. The nitrogenase-containing cells possessed carboxysomes that were rich in ribulose-1,5-bisphosphate carboxylase/oxygenase and phycoerythrin, a light harvesting pigment of PS II. This indicates that these cells had a capacity for both N2 fixation and photosynthesis. The significance of the localization pattern for dinitrogenase reductase is discussed in the context of N2 fixation in Symploca PCC 8002.

It is becoming increasingly apparent that the long-distance signalling associated with many developmental processes is complex and that novel hormone-like signals may play substantial roles. The past decades have seen several substances (e.g. brassinosteroids, systemin and other polypeptides, mevalonic and jasmonic acids, polyamines, oligosaccharides, flavonoids, and quinones) vie for a place among the classical plant hormones (e.g. Spaink, 1996). Recent microinjection and grafting studies have also shown that RNA may act as a long-distance signal (Jorgensen et al., 1998; Xoconostle-Cázares et al., 1999). In this issue, Hannah et al. describe long-distance signalling and the regulation of root–shoot partitioning in dwarf lethal or dosage-dependent lethal (DL) mutants of common bean (Shii et al., 1980, 1981), and present evidence indicating that substances in addition to classical plant hormones (e.g. cytokinins) may be involved.

As in the report by Hannah et al., much of the evidence for roles of unidentified long-distance signals in the control of plant development is indirect. The possibility that a small number of long-distance signals might control a multitude of developmental processes arises through the potential for differences in tissue sensitivity, fluctuations in hormone levels and differences in the nature of responses of different tissues to the same hormone. Consequently, particular hormones may influence numerous processes seemingly simultaneously, yet independently. Even so, long-distance signalling is involved in processes as diverse as root–shoot balance, senescence, branching, flowering, nodulation, stress responses and nutrient uptake. Through comparison of even a few different developmental processes, progress can be made to reveal the true complexity of plant development. Using this approach it is also clear that many unknown signals may be involved.

A detailed study was carried out to obtain information on the relative rates of NH4+ and NO3−-N uptake and assimilation by perennial ryegrass (Lolium perenne L.), when grown in soil and supplied with different ratios of NH4+ and NO3−-N. Following two preliminary defoliations, plants were supplied with 15N-(differentially) labelled NH4+ and NO3−-N in three different ratios (20[ratio ]80, 50[ratio ]50 and 80[ratio ]20), followed by sequential destructive harvests of shoots and roots taken at four points during a 35-d regrowth period. When supplied with equimolar concentrations of NH4+ and NO3−-N, perennial ryegrass absorbed both forms of nitrogen at almost identical rates; and even when the two N forms were supplied in widely disproportionate concentrations, both forms appear to have been absorbed at equal rates, at least until the supply of the minor nitrogen component in each treatment was almost exhausted, i.e. by day 3 of regrowth. It is suggested that this matching of NH4+ and NO3−-N absorption rates by plants maximizes the advantages and minimizes the disadvantages associated with the exclusive use of either form of nitrogen, and thereby optimizes the potential for vegetative and reproductive regeneration. Fifteen days after the application of 15N-labelled fertilizer, shoot and root yields differed little between treatments. However, plants grown with NH4+ and NO3−-N at a ratio of 50[ratio ]50 had accumulated significantly more root (soluble) carbohydrate than those in the other two treatments. Consequently, when subjected to defoliation at day 15, these former plants were clearly in the best position to re-mobilize root carbohydrate and initiate shoot regeneration. However, further work is needed in order to determine whether or not this finding holds true for plants subjected to a multiple (bi- to tri-weekly) defoliation regime, e.g. in a grazing situation.

One alkaline invertase and two acid invertase activities were detected in the shoots of etiolated rice (Oryza sativa) seedlings. The alkaline invertase (AIT) was purified to homogeneity through steps of ammonium sulphate fractionation, concanavalin A-Sepharose affinity chromatography (non-retained), DEAE-Sephacel chromatography and preparative electrophoresis. The pH optimum of AIT was 7.0 and the molecular mass, determined by gel filtration, was 240 kDa. It is apparently a homotetrameric enzyme (subunit molecular mass 60 kDa). The isoelectric point was 4.4 by isoelectric focusing. The best substrate of the enzyme was sucrose, with a Km of 2.53 mM. The enzyme also hydrolysed raffinose, but not maltose or lactose, so it is a β-D-fructofuranosidase. It gave negative glycoprotein staining. Of the hydrolysis products, fructose was a competitive inhibitor and glucose was a non-competitive inhibitor. Treatment with an alkaline phosphatase could activate AIT, whereas other proteins such as BSA, concanavalin A and urease had no effect on the enzyme activity. The enzyme activity was inhibited by Tris, thiol reagents and heavy metal ions.

Eragrostis pilosa (Linn.) P Beauv., a C4 grass native to east Africa, was grown at both ambient (350 μmol mol−1 and elevated (700 μmol mol−1) CO2 in either the presence or absence of the obligate, root hemi-parasite Striga hermonthica (Del.) Benth. Biomass of infected grasses was only 50% that of uninfected grasses at both CO2 concentrations, with stems and reproductive tissues of infected plants being most severely affected. By contrast, CO2 concentration had no effect on growth of E. pilosa, although rates of photosynthesis were enhanced by 30–40% at elevated CO2. Infection with S. hermonthica did not affect either rates of photosynthesis or leaf areas of E. pilosa, but did bring about an increase in root[ratio ]shoot ratio, leaf nitrogen and phosphorus concentration and a decline in leaf starch concentration at both ambient and elevated CO2. Striga hermonthica had higher rates of photosynthesis and shoot concentrations of soluble sugars at elevated CO2, but there was no difference in biomass relative to ambient grown plants. Both infection and growth at elevated CO2 resulted in an increase in the Δ13C value of leaf tissue of E. pilosa, with the CO2 effect being greater. The proportion of host-derived carbon in parasite tissue, as determined from δ13C values, was 27% and 39% in ambient and elevated CO2 grown plants, respectively. In conclusion, infection with S. hermonthica limited growth of E. pilosa, and this limitation was not removed or alleviated by growing the association at elevated CO2.

Maize (Zea mays L.) plants parasitized by the root hemi-parasitic angiosperm, Striga hermonthica (Del.) Benth., consistently display a range of symptoms similar to those found in droughted plants. The mechanisms by which these changes occur are largely unknown. However, S. hermonthica has unusually high rates of transpiration, and stomata which are relatively insensitive to water deficit. Consequently, it has often been suggested that the parasite might cause a severe depletion of the available water in the host's rooting zone. To determine whether the lower stomatal conductance and retarded growth of infected plants could be a result of parasite-induced water deficit, we have monitored the matric potential of the growth medium, water use, growth and stomatal conductance of infected vs. uninfected maize plants.

Host plant height and stomatal conductance of parasitized plants were significantly lower than those of control plants from 31 or 37 d after planting (d.a.p.) respectively. However, there was no indication of an increase in the rate of water depletion in the rooting zone of infected plants until approx. 63 d into the parasitic association. In fact, from 39 until 59 d.a.p. infected plants used less water than uninfected control plants, probably the result of the plants having fewer expanded leaves during part of this period, combined with the lower stomatal conductance exhibited by the infected plants from day 37 onwards. Leaf RWC of infected plants was unchanged in comparison with that of uninfected plants, therefore the change in stomatal conductance was not a response to dehydration of the leaf tissue. Our results indicate that parasitism by S. hermonthica does not cause an increase in water uptake/use in the host until well after most of the symptoms of infection have become fully established. It is highly unlikely, therefore, that the observed effects on the host are primarily due to soil water deficit.

Lipids were extracted from needles of Norway spruce plants of different provenances. The plants, seeds of which were originally collected from all over Europe, were grown at two sites in England at the same longitude but one 350 km north of the other. Significant correlations were observed between monogalactosyl diglyceride (MGDG) and digalactosyl diglyceride (DGDG) ratios on the one hand and, on the other, the lowest extreme winter temperature at the original site of seed collection (EWTS).

In the case of lipid changes, however, environmental differences due to site were more important than the inherent genotypic characteristics of the planted trees while provenances with the lowest EWTS values had more MGDG than DGDG. This implies that existing winter hardiness under mild climatic conditions does not come from changes in this ratio although DGDG formation from MGDG is known to be an important feature of winter hardening in Norway spruce elsewhere in Europe. However, closer analysis of the fatty acids in lipids from provenances at individual sites did reveal significant correlations between those provenances with low EWTS values and certain molar amounts or ratios of fatty acids in DGDG, some of which could be traced to genotypic expression. These partly involved the unusual Δ5 desaturations which are mainly confined to the fatty acids of those conifers like Norway spruce that harden to very low temperatures.

Plant species that persist during succession, from the colonization to the stabilization stages, face major environmental changes. Such changes are believed to have significant effects on species performance. In subarctic coastal dune systems, Leymus mollis colonizes the embryo dunes, on the upper limit of the beach. It reaches its maximum density on the foredune, but also grows on older, stabilized ridges. This paper reports on the phenotypic variations of some ecophysiological traits associated with the persistence of L. mollis on a dune system on the east coast of Hudson Bay (northern Quebec). Leymus mollis ramets tend to have a lower net carbon assimilation rate and water use efficiency, and a higher substomatal CO2 concentration on the stabilized dune than on the foredune. However, these physiological differences cannot be explained by differences in leaf morphology or nitrogen content. Under controlled conditions, ecophysiological differences observed in the field disappear, suggesting that these are not genetic but determined by environmental changes along the foredune-stabilized dune gradient. We propose that higher net carbon assimilation rate on the foredune might be related to higher sink strength in relation to the growth-stimulating effect of sand burial.

Compatible pollination of Brassica napus necessitates pollen hydration, pollen germination and growth of the pollen tube through the loosened walls of stigmatic papillar cells, whereas self-incompatible (SI) pollinations fail at one of these stages. Analyses of the early stages of pollination show that at high (but not low) relative humidities, both compatible and SI pollen hydrates, but SI germination is reduced and the rare pollen tubes generally fail to penetrate the papillar walls, although there is some wall loosening. Inside the papillae, both compatible and SI interactions may induce the formation of callose, but there is no evidence for a major accumulation of cytoplasm or secretory vesicles in the vicinity of the pollen tubes and neither microtubule nor F-actin patterns re-arrange in this zone. These observations indicate that the source of the wall-loosening enzymes is probably the pollen tube or pollen coat, and that the common cellular responses of plants to attempted invasions have become suppressed in the papilla–pollen tube interaction.

Anti-transpiration and anti-growth activities of xylem sap were studied for maize, sunflower, cotton and castor bean, which were subjected to several stress treatments: soil drying, flooding and salinity. All samples of xylem sap showed an increased concentration of proteins when plants were either soil-dried, salt-treated or flooded. The protein transportation flux in xylem sap, i.e. the product of protein concentration and volume flux, was also increased as a result. Electrophoresis using SDS–PAGE analysis showed that xylem proteins were of variable size. When samples of xylem sap were used to feed detached leaves or shoots in a bioassay system for their anti-transpiration and anti-growth activities, such xylem proteins in their original concentrations apparently showed neither direct inhibition nor enhanced effects of ABA. All samples of xylem sap from plants subjected to different stresses showed anti-transpiration and anti-growth activities that could be fully accounted for by their ABA concentration. In cases where such activities were more than could be explained by the ABA concentrations, filtration with a 0·20 μm microfilter removed the discrepancies.

Both sensitive and tolerant clones of aspen (Populus tremuloides) were exposed to ozone using four different exposure regimes under controlled environmental conditions. Based on data on ambient ozone from 10 cities in the USA, three treatments of 4-wk exposure to the same SUM06 (an accumulation of hourly O3 concentrations greater than 0.06 ml l−1) were constructed. The regimes allowed us to investigate: (a) the importance of long (3 wk, treatment 1) versus short (1 wk, treatment 2) duration of regimes with high peaks; (b) the effect of treatments with variable peak occurrence (treatments 1 and 2) versus uniform peak occurrence (treatment 3) during the exposure period. Nonfumigated control plants were maintained at ozone concentrations <10 nl l−1. Bifacial black necrosis, a typical symptom of ozone injury on aspen leaves, occurred on both clones after 2 wk exposure. Up to 60% of the leaves on the sensitive clone were injured, with an average of 6% of total leaf area injured. In the tolerant clone only 10% of the leaves were injured, with less than 1% of the total leaf area symptomatic. The severity of injury was consistently greatest in treatment 2, followed by treatments 1 and 3, respectively. The interval between peak exposures was less important than the occurrence of peaks versus a stable maximum concentration. Premature leaf abscission occurred in the sensitive clone. Measures of gas exchange demonstrated reduced photosynthesis under ozone fumigation, but exposure regime was not a significant factor. Concentrations of two antioxidants, ascorbic acid and glutathione, were almost always greater in the resistant than in the sensitive clone, but the differences were not statistically significant. The levels of these antioxidants in aspen leaves did not change with ozone fumigation or leaf age.

The incorporation of [35S]sulphur in thiophenes by Tagetes patula roots was used as a model to study the regulation of secondary metabolism with a limited supply of substrate. Growth and thiophene accumulation were measured in root cultures incubated at various sulphate concentrations in the medium. A 20-fold to 40-fold reduction in the sulphate concentration did not affect elongation growth, branching and biomass production within 14 d but decreased the thiophene level to 25–50% of the control in the same period. The reduction in thiophene content was found to result from a decline in biosynthetic capacity of 80–95% after 8 d. This capacity was restored when roots were transferred to standard medium. The restoration took more than 24 h and was suppressed by cordycepin, an inhibitor of mRNA processing. It is concluded that the rate of thiophene synthesis is regulated by a control mechanism that reacts to the availability of sulphate to the roots.

Differences between individuals collected from sun and shade populations could result from either somatic or genetic differences, particularly in populations of perennial plants. Our objective in this study was to separate somatic from genetic differences. We collected Anthoxanthum odoratum and Plantago lanceolata from sun and shade populations and made measurements on both vegetatively propagated clones and seed progeny from each clone. The parental populations differed in a wide range of physiological and morphological traits. However, only photosynthetic capacity was significantly different between both the original sun and shade populations and their seed progeny. In both species, plants from the sun population had higher photosynthetic capacities than those from the shade population when grown in a common environment. This demonstrates that there was genetic differentiation between the sun and shade populations. Photosynthetic capacity of parents and offspring also differed, suggesting a somatic effect. Since many of the original clones were virus-infected, but all but one of the offspring were virus-free, this might have been a result of virus infection. However, in spite of the fact that the parents and offspring clones were propagated vegetatively so that the plants were at the same developmental stage at the time of measurement, we cannot rule out the possibility that differences in age of cell lines could also have been a factor.

The relative rates of ammonium and nitrate-N uptake and assimilation by creeping bent (Agrostis stolonifera), were investigated for plants grown in soil and supplied with three different ratios of ammonium and nitrate-N. Following two preliminary defoliations, plants were supplied with the equivalent of 150 kg N ha−1, given as 15N-(differentially) labelled NH4+ and NO3−-N in three different ratios (20:80, 50:50 and 80:20), followed by sequential destructive harvests of shoots and roots at four points during a 35-d regrowth period. Maximum use of labelled nitrogen and ‘exhaustion’ of soil mineral nitrogen reserves occurred much earlier when plants were supplied with half or more of their nitrogen as ammonium, than occurred when they were supplied predominately with nitrate-N. The lack of consistency in the patterns of ammonium and nitrate-N absorption, however, implied that the plants had no specific preference for either nitrogen form. Supplying plants with different combinations of ammonium and nitrate produced distinctive differences in plant morphology. In the high nitrate treatment, plants preferentially partitioned resources into shoot and stolon formation, whereas in the high ammonium treatment, resources were preferentially partitioned into root production. These changes in plant morphology might be adaptations to aid species survival in environments associated with a predominance of either nitrogen form.

Several temporal and spatial scales have been considered during the Symposium, and the presentations by Wallenda & Kottke (1998) and by Norby (1998) represent two extreme points in a wide range. They both deal with short and long-term impacts on the local as well as the global scale. I present here a few additional thoughts, and a subject that both papers have touched only slightly will be expanded: the differential impact of different forms of nitrogen deposition.
