Three lepidopteran cereal stemborers, Chilo partellus (Swinhoe) (Crambidae), Busseola fusca Fuller, and Sesamia calamistis Hampson (Noctuidae) were collected from maize and sorghum in Ethiopia. The noctuid stemborers are indigenous to Africa while C. partellus is an introduced species from Asia. In 1999, the Asian stemborer parasitoid, Cotesia flavipes Cameron (Braconidae) was found to be widespread in Ethiopia, even though it had never been released in the country. In addition to attacking Chilo partellus, Cotesia flavipes was reared from B. fusca and S. calamistis. The origin of C. flavipes in Ethiopia may have been Somalia where it was released in 1997 near the border with eastern Ethiopia. Percent parasitism of borers by C. flavipes was higher in eastern Ethiopia than other surveyed regions, and parasitism was higher in 2000 than 1999. Parasitism was higher when cereals were intercropped with other plants and when wild grass hosts of stemborers were present.

The establishment of the predatory histerid beetle, Teretrius nigrescens Lewis, following its field release in south eastern Kenya in 1992, and its impact on populations of the larger grain borer, Prostephanus truncatus (Horn), in woodland habitats is evaluated. Pheromone trap catch data show a single strong peak of P. truncatus from November to January, coincident with the short rains. Comparisons of P. truncatus pheromone trap catches before and after predator establishment, and in areas with and without T. nigrescens, show a sustained decline of over 80% in P. truncatus abundance over a 5-year period, following the establishment T. nigrescens. Concurrent laboratory studies of the impact of T. nigrescens on populations of P. truncatus raised on a wood substrate showed a 77% reduction in the growth of the pest population. These results are compared with published findings of the impact of T. nigrescens as a biological control agent of P. truncatus in west Africa.

A high diversity library of recombinant human antibodies was selected on complex antigen mixtures from midguts of female Anopheles gambiae Giles. The library of phage-displayed single chain variable region fragment constructs, derived from β-lymphocyte mRNA of naïve human donors, was repeatedly selected and reamplified on the insoluble fraction of midgut homogenates. Five rounds of panning yielded only one midgut-specific clone, which predominated the resulting antibody panel. In A. gambiae, the epitope was found throughout the tissues of females but was absent from the midgut of males. The cognate antigen proved to be detergent soluble but very sensitive to denaturation and could not be isolated or identified by Western blot of native electrophoresis gels or by immunoprecipitation. Nevertheless, immunohistology revealed that this sex-specific epitope is associated with the lumenal side of the midgut. Severe bottlenecking may limit the utility of phage display selection from naïve libraries for generating diverse panels of antibodies against complex mixtures of antigens from insect tissues. These results suggest that the selection of sufficiently diverse antibody panels, from which mosquitocidal or malaria transmission-blocking antibodies can be isolated, may require improved selection methods or specifically enriched pre-immunized libraries.

The distributions of insecticide-treated cattle from sites in Tanzania and Zimbabwe were assessed from interviews with livestock owners, analysis of secondary livestock data and mapping technologies. The time-course of tsetse control operations at these sites were then simulated using a mathematical model that assumed diffusive movement and logistic growth in fly populations. A simulation of a tsetse control operation in Mudzi district, north-east Zimbabwe, was in accord with observations that the use of insecticide-treated cattle was unable to prevent substantial re-invasion of tsetse from Mozambique, consequent on the patchy distribution of cattle. The simulation was also consistent with the observed efficacy of a 10-km wide barrier of insecticide-treated targets deployed evenly at 4 km−2. Simulation of a control operation on Mkwaja Ranch in Tanzania was in accord with the observation that the use of insecticide-treated cattle reduced the tsetse population on the ranch by c. 90%. Insecticide-treated cattle were used to better effect in the Kagera Region of Tanzania. Simulation of this operation predicts that the deployment of 35,000 treated cattle in the area would result in > 99% control of the tsetse population, consistent with the observed decline, by 1–2 orders of magnitude, in cases of trypanosomiasis in the region. The greater success of the Kagera operation was due to the size and shape of the treated area and, particularly, to the restriction of re-invasion to 20% of the perimeter, compared with > 80% on Mkwaja. Simulation was used to assess how tsetse control could have been improved at Mkwaja. The results suggest that splitting herds into smaller, more numerous, units could have achieved some improvement but, in general, the disease problem would not have been solved by the use of insecticide-treated cattle alone. Only by deploying odour-baited targets in ungrazed areas, or in a 1–3-km barrier around the ranch, could substantially better control (99–99.9%) have been achieved. Sensitivity analyses of the Mkwaja simulation showed that the general conclusions were robust to assumptions regarding cattle distribution and the rates of fly movement and growth. Properly managed and appropriately applied insecticide-treated baits are powerful weapons for tsetse control but should not be used without regard to potential levels of re-invasion, consequent largely on considerations of the size and shape of the treatment area and the density and distribution of the baits.

The exploration of sand-filled arenas by workers of an entire colony of the Australian, subterranean foraging, tree-nesting termite, Coptotermes frenchi Hill was investigated under laboratory conditions. The first experiment tested whether termite exploration of sand was influenced by the presence of gaps or objects in the sand. Gaps and objects were chosen to represent soil heterogeneity in the urban environment: gaps to represent tunnels dug by other animals, perspex strips to represent cables and pipes, and wood strips to represent roots. Termites always chose to explore gaps thoroughly before they began tunnelling in the sand. Significantly more and longer tunnels were excavated from the end of gaps at the far end of the arenas, and relatively little tunnelling occurred around and along objects. Termite density was significantly greater around and along wood compared with perspex blocks. The second experiment tested whether termite exploratory tunnelling was influenced by soil moisture. The termites tunnelled slowly in dry sand, but after discovering a patch of wet sand, increased tunnelling five-fold until it was completely explored, after which activity declined. Energy and water conservation may be behind these patterns of exploratory tunnelling as well as those seen in large field studies, but caution is urged when interpreting small scale laboratory experiments to explain large scale field data.

Biological and population parameters of the predatory bugs Macrolophus costalis Fieber and Macrolophus pygmaeus Rambur preying on the tobacco aphid Myzus persicae (Sulzer) were examined. Tobacco was used as host plant and all experiments were carried out at 23°C and L16:D8. In M. costalis, the developmental time for eggs and the total duration of nymphal instars was 13.1 and 22.2 days and in M. pygmaeus 13.0 and 20.2 days, respectively. Both species completed their nymphal development feeding only on plant juices although they required significantly more time to attain adulthood than when feeding on prey. The longevity of M. costalis females and the preoviposition period were 49.9 and 5.6 days and those of M. pygmaeus were 50.3 and 6.3 days, respectively. Adult males showed a higher longevity than females in both species. The intrinsic rate of increase and the mean total number of eggs laid were 0.0644 and 121 in M. costalis and 0.0615 and 104 in M. pygmaeus, respectively. Mean total aphid consumption by nymphs of M. costalis was 61 aphids, about twice that of M. pygmaeus (37 aphids). Total aphid consumption by a male and female pair of M. costalis and M. pygmaeus until the death of the female was 244 and 285 aphids, respectively. The results of the study are discussed in relation to the impact of the two predatory bugs on tobacco aphid populations.

In this study, the question of whether Childers canegrub, Antitrogus parvulus (Britton) overwinters in the subsoil was addressed. Irrigated fields of sugarcane were sampled during a 2-year period near Bundaberg in southern Queensland. Antitrogus parvulus overwintered as second and third instars at each of three sites. During autumn and winter third instars of different allochronic (separated in age by 12 months) populations occurred together and could not be readily separated. Field-collected third instars were reared on ryegrass and separated into two age groups based on the date of pupation. Third instars in the first year of their life cycle (young third instars) remained at shallow depth (100–200 mm) and did not overwinter in the subsoil as once thought. Minimum temperatures during winter were 13–16°C and did not prevent young third instars from feeding and gaining weight. Third instars in their second and final year moved downwards from late summer and pupated in the subsoil at 293–425 mm in spring. General phenology was as previously reported with first instar larvae occurring from January until April, second instars from January until November and third instar larvae throughout the year. Prepupae and pupae were found between October and December and adults occurred in soil during November and January. Batches of eggs occurred at a mean depth of 350 mm. First and second instars occurred predominantly at relatively shallow (100–200 mm) depths in the soil profile. All stages tended to be most common under rows of sugarcane rather than in the interrow.

Multivariate morphometric analysis (method of canonical variates) was used to compare 38 parthenogenetic lineages and three field collections of aphids of the Myzus persicae (Sulzer) group sampled on peach and tobacco in the Caserta region of southern Italy. Comparisons were also made with the morphology of lineages collected on peach in Lehonia, in central eastern Greece away from tobacco-growing regions, and on tobacco in Naphplion, in southern Greece. The lineages were measured after parthenogenetic rearing for one to three generations on the same host (potato) under constant conditions. As in previous work, the multivariate morphometric approach separated the aphids from peach from those feeding on tobacco. The life cycle category of the lineages was also examined. Almost all the lineages from peach in Caserta were holocyclic, yet the lineages from tobacco in the same region were unable to produce sexual morphs. The results suggest that tobacco-adapted and non-tobacco-adapted forms co-exist in the same region in southern Italy, not only because they colonize different host plants, but also because they have different life cycles. This is in complete contrast to the situation in peach-growing areas of northern Greece, and shows that the ecology and population structure of M. persicae is different in neighbouring counties of the Mediterranean area, even where climatic conditions and cultivated crops are similar.

A description of Anopheles (Cellia) irenicus Schmidt, sp.n. (formerly A. farauti No. 7) is provided. This species is one of six recorded from the Solomon Islands within the A. punctulatus group, which contains the major vectors of the causative agents of malaria and lymphatic filariasis in the southwest Pacific. Morphological markers are described for adult females, fourth-instar larvae and pupae that identify most specimens of A. irenicus. Keys are presented to distinguish members of the A. punctulatus group in the Solomon Islands.

Field experiments were performed in artificial ponds to evaluate how the density of predatory diving beetles (Dytiscidae) would affect the population levels of mosquito larvae (Culicidae). Mosquitoes colonizing the ponds were predominantly species of the genus Culex. In 2000, most of the dytiscids colonizing the ponds were small (Hydroporus spp.), and these predators had no impact on the size of larval mosquito populations, not even in ponds with added dytiscids. In 2001, larger beetles (Ilybius, Rhantus, and Agabus spp.) were more common, and there were significantly fewer mosquito larvae in ponds with the highest numbers of dytiscids. There was a negative correlation between numbers of diving beetles in the ponds and the mean body length of mosquito larvae. In neither year could dytiscid densities be maintained above a certain level owing to emigration. In laboratory tests, there were marked differences between three common dytiscid species in regard to preferences for Daphnia and Culex species as prey: Colymbetes paykulli Erichson chose mosquito larvae more often, whereas both Ilybius ater (De Geer) and I. fuliginosus (Fabricius) preferred Daphnia spp. All of the tested dytiscids consumed large numbers of prey. Since some dytiscid species can efficiently decrease populations of mosquito larvae, they are probably important in the natural control of these dipterans.

Chrysophtharta agricola (Chapuis) is a pest of commercial eucalypt plantations in Tasmania and Victoria. Vagility of pest populations may result in difficulty predicting temporal and spatial pest outbreaks, and influence genetic resistance to chemical control. Gene flow in this pest species was estimated to assess predicability of attack, the potential efficacy of natural enemies, and the likelihood of resistance build-up. Ten geographic populations of C. agricola (six from Tasmania, one from the Australian Capital Territory, one from New South Wales and two from Victoria) were examined for genetic variation and gene flow using cellulose acetate allozyme electrophoresis. Six enzyme systems (PGI, PGD, PGM, IDH, HEX and MPI) were consistently polymorphic and scorable and were used to quantify estimated gene flow between populations. FST values and analysis of molecular variance indicated that gene flow was restricted between populations. Chrysophtharta agricola exhibited high levels of heterozygosity, probably because of high allelic diversity, and because all loci examined were polymorphic. The southern-most population was the most genetically different to other Tasmanian populations, and may also have been the most recently colonized. Limited gene flow implies that outbreaks of C. agricola should be spatially predictable and populations susceptible to control by natural enemies. Our results also imply that genetic resistance to chemical control may occur under frequent application of insecticide. However, testing population movement between plantations and native forest also needs to be conducted to assess gene flow between forest types.

The responses of male and female Lutzomyia longipalpis (Lutz & Neiva) to different wavelengths of light was tested by presenting the sandflies with two light sources simultaneously, a series of test wavelengths between 350–670 nm and a 400 nm control. To test whether L. longipalpis could discriminate between the test and control, three sets of experiments were carried out in which the test wavelengths were presented at higher, equivalent or lower intensity than the control. In all three experiments, ultra-violet (350 nm) and blue-green-yellow (490–546 nm) light was more attractive to L. longipalpis than the control wavelength. However, at low intensity, UV was less attractive, than equivalent or higher intensity UV light. At intensities equivalent to or higher than the control wavelength, ultra-violet light was more attractive than blue-green. Furthermore, at low intensity, green-yellow (546 nm) light was more attractive to males whereas blue-green (490 nm) was more attractive to females. Blue-violet (400 nm) and orange-red (600–670 nm) light were least attractive in all three sets of experiments. Response function experiments indicated that the responses were dependent on both intensity and wavelength and that therefore more than one photoreceptor must be involved in the response. The results indicated that L. longipalpis can discriminate between different wavelengths at different intensities and thus have true colour vision. It also suggests that L. longipalpis may be able to navigate at dusk or under moonlight or starlight conditions using light in the blue-green-yellow part of the spectrum. The difference in response of males and females to light in this region is interesting and may indicate the different ecology of the sexes at night. Overall, these results may have important implications for sandfly trap design.

The impact of predation by the generalist carabid beetle Pterostichus melanarius (Illiger) on populations of the field slug Deroceras reticulatum (Müller), and the effects of prey size on the predator–prey interaction, were measured under semi-field conditions. It was hypothesized that environmental heterogeneity would lead to very different patterns of comparative mortality than might be deduced from size choice experiments conducted in the laboratory. Results from outdoor mini-plots, emulating conditions in a field of wheat, demonstrated that P. melanarius significantly reduced numbers of slugs from all size classes, with no apparent preferences. This was in marked contrast to results from earlier laboratory studies, where this beetle fed preferentially on the smallest slugs. The slugs in the mini-plots ranged in size from 2–100 mg and the numbers in the mini-plot reflected the size frequency distribution in the field. Beetles in mini-plots containing high densities of slugs increased significantly in weight, in contrast to beetles in mini-plots with low slug density or no added slugs, which did not. Enzyme-linked immunosorbent assays (ELISA), using anti-slug monoclonal antibodies, showed that where there was a higher density of slugs there was more slug protein in the guts of the beetles. It was concluded that environmental heterogeneity probably provided a greater number and diversity of refugia for smaller than for larger slugs, counteracting laboratory-measured size preferences measured in arenas without refugia. These results have implications for a range of ecological studies involving inter- and intra-specific prey size choice, and emphasize the dangers of extrapolating from the laboratory to the field.

Populations of many phloem-feeding aphid species in temperate regions increase exponentially in early summer and then ‘disappear’, usually over a time-scale of a few days, in July. To understand these dynamics, empirical investigation of the causes and modelling of the processes underlying population change are required. Numbers of the aphids Myzus persicae (Sulzer) and Macrosiphum euphorbiae (Thomas), monitored over three years in commercial potato fields in the UK, increased to a maximum of 2–2.5 per leaflet on 16 July in 1999 and 2001, and then declined to < 0.25 per leaflet by 26 July. In 2000, aphid numbers remained very low (< 0.25 per leaflet) throughout the season. The onset of the crash in aphid numbers (16–19 July in 1999 and 2001) was consistently associated with changes in the phloem amino acid composition of potato leaflets. Natural enemies, including syrphids, parasitoids, coccinellids, chrysopids and entomopathogenic fungi, increased in abundance throughout the sampling period. The incidence of winged emigrant aphids prior to the crash was low (< 10%). Experimental manipulation during 2001 demonstrated that, during the crash period, the fecundity of aphids (caged on leaves to exclude natural enemies) was depressed by 25–45% relative to earlier in the season, and that presence of natural enemies reduced aphid numbers by up to 68%. Using these data, an excitable medium model was constructed, which provided a robust description of aphid population dynamics in terms of plant development-induced changes in aphid fecundity and temporal change in natural enemy pressure.

The susceptibility to several insecticides of 16 and 8 strains of Myzus persicae Sulzer and Aphis gossypii Glover, respectively, received from different European countries in 2001 was investigated. Most of the strains were derived from places known for their aphid resistance problems to conventional insecticides before imidacloprid was introduced. In many regions and agronomic cropping systems imidacloprid has been an essential part of aphid control strategies for a decade, and therefore the susceptibility of aphid populations to imidacloprid using FAO-dip tests and diagnostic concentrations in a leaf-dip bioassay was checked. Additional insecticides tested were cyfluthrin (chemical class: pyrethroid), pirimicarb (carbamate), methamidophos and oxydemeton-methyl (organophosphates). Diagnostic concentrations (LC99-values of reference strains) for each insecticide were established by dose response analysis using a new leaf-disc dip bioassay format in 6-well tissue culture plates. Virtually no resistance to imidacloprid in any of the field-derived populations of M. persicae and A. gossypii was detected. In contrast, strong resistance was found to pirimicarb and oxydemeton-methyl, and to a lesser extent also to cyfluthrin. Two strains of A. gossypii exhibited reduced susceptibility to imidacloprid when tested directly after collection. However, after maintaining them for six weeks in the laboratory, the aphids were as susceptible as the reference strain. The diagnostic concentration of methamidophos did not reveal any resistance in M. persicae, but did so in four strains of A. gossypii.

Mosquitoes of the Anopheles maculipennis complex were collected in nine provinces of Iran (Esfahan, Fars, Gilan, Golestan, Kohkiluyeh va Boyerahmad, Mazandaran, Tehran, Azarbaijan-e Gharbi and Zanjan) between June 1983 and September 2002. The nuclear rDNA ITS2 sequences of 86 specimens were compared with those of seven species of the complex available in GenBank. Three genetically distinct species of the complex were distinguished: A. maculipennis Meigen, A. sacharovi Favre and a previously unrecognized species. The last species is most similar to, but clearly distinct from, A. martinius Shingarev and A. sacharovi. The taxonomy of A. martinius and A. sacharovi is critically reviewed, and justification is provided for formally recognizing the third species as Anopheles persiensissp.n. The new species is the first culicid to be characterized and named principally on the basis of DNA evidence. Anopheles persiensis was collected only in the northern Caspian Sea littoral provinces of Gilan and Mazandaran, and it seems likely that this species could be responsible for malaria transmission in this region that was previously attributed to A. maculipennis. A species-specific RFLP–PCR assay based on ITS2 sequences was developed to facilitate further studies of the three species in Iran.

Forty-one accessions of wild and cultivated wheats belonging to 19 Triticum species were tested in the field for resistance to three species of aphids, Rhopalosiphum padi Linnaeus, Sitobion avenae Fabricius and Schizaphis graminum Rondani. Antibiotic resistance was estimated by the increase in biomass of aphids over 21 days on adult plants. Overall resistance was estimated by the plant biomass lost due to aphid infestation. All three species of aphids survived and reproduced on all wheats, and reduced spike biomass compared to uninfested controls. The level of antibiosis varied among wheat species and among accessions, with accessions from three, five and one species showing antibiosis to R. padi, S. avenae and S. graminum, respectively. Overall resistance to the three aphid species was observed in five to seven accessions per aphid species. Resistance was usually specific to one aphid species. The frequency of accessions with antibiosis or overall resistance was associated with the ploidy level of the plant species. Except for overall resistance to R. padi, resistance was highest for diploid species and lowest for hexaploid species. No consistent relationship between resistance and level of domestication was detected. Accessions of the wild wheats, Triticum boeoticum Bois, Triticum tauschii (Coss.) Schmal. and Triticum araraticum Jakubz. exhibited high levels of resistance to aphids, as did Triticum monococcum L. which is derived from T. boeoticum. Nevertheless, individual susceptible or resistant accessions occurred at all levels within the evolutionary tree of wheat.

The performance of eighteen clones of Myzus persicae (Sulzer) on pepper and tobacco plants at 20°C and L16:D8 and the choice of young adult apterae between tobacco and pepper leaf-discs were examined. The clones were collected from weeds and peach in two tobacco-growing regions: Katerini, northern Greece and Karditsa, central Greece (only from weeds) and from Lehonia, central eastern Greece where tobacco is not cultivated. All clones did well on both hosts. However, the analysis of data revealed a significant effect of ‘region / host plant origin’ on aphid performance. The mean values of adult weight, intrinsic rate of increase and fecundity of the clones collected in Lehonia and reared on tobacco were significantly lower than the observed values for clones from Katerini and Karditsa. Aphids from Lehonia had significantly higher mean values for developmental time on tobacco than clones from the other regions whereas the opposite was observed when aphids were reared on pepper. Aphids collected in Lehonia performed better on pepper than those originating from the tobacco-growing regions. A choice test revealed differences among the clones originating from different regions. Fifty three percent and 43% of aphids from weeds and peach from Lehonia, respectively, chose pepper. By comparison 41.5% and 40.0% of aphids from peach and weeds from Katerini, respectively and 49.5% of aphids from Karditsa preferred tobacco. The results are discussed in relation to host specialization in M. persicae.

An Australian gall-inducing eulophid, Ophelimus eucalypti (Gahan) was first recorded on the foliage of Eucalyptus botryoides after it invaded New Zealand in 1987. It has spread throughout the eucalypt plantations in the North Island and in the northern parts of the South Island affecting several species of Eucalyptus in the section Transversaria (subgenus Symphyomyrtus). Because gall-inducing insects usually have extremely narrow host ranges, O. eucalypti that induces galls on E. saligna and E. botryoides is currently recognized as a biotype, O. eucalypti (Transversaria). Heavily galled leaves abscise from the plant. Repeated defoliation led to widespread die-back of susceptible eucalypt species in the 1990s. Female larvae of O. eucalypti induce circular, protruding galls on the leaves of E. botryoides and E. saligna, whereas the males induce pit galls on the same species. The biology of O. eucalypti females and the development of their galls are described. Adult female O. eucalypti antennate the leaf surface before inserting the ovipositor (otherwise concealed within the metasomal apex) into the young host leaf. The egg is inserted at approximately 45° and discharged between differentiating palisade cells. Callus-type cells surround the egg chamber, but cytologically specialized nutritive cells appear once the egg hatches and the larva begins to feed. The gall also differentiates a multi-layered sclerenchymatous tissue around the nutritive tissue. After feeding for many months, the larva pupates and the active nutritive tissue degenerates. The adult wasp emerges after cutting an exit hole through to the outside of the gall. Abscission of heavily galled leaves results in widespread defoliation and loss of growth and vigour in susceptible trees in New Zealand.

Thirty Reticulitermes flavipes (Kollar) colonies established by alates collected from two separate field sites were raised in the laboratory for eight years. Twenty-one of the colonies were founded by alates from one field source and nine from another, providing demographic data from two unrelated parental lineages. Colony totals ranged from 3620 to 11641 individuals, with no significant difference in size between lineages. Soldier caste proportion of the colony total and mean wet weights for workers, soldiers and kings were significantly different between the two lineages. This suggests that at least a portion of the variability observed in caste ratios and body size may be heritable. One founding reproductive had died in five of the colonies (17%); none lost both parents. The queenless colonies contained exclusively female replacement reproductives (neotenics); the kingless colony contained a female-skewed mixture of male and female neotenics. All the nests that lost a founding parent contained significantly more pre-alate nymphs than the nests with both a king and a queen. Comparisons with published reports of ontogenetic patterns in other termites and social insects are discussed.
