The concentrations of organotin (OT) compounds, butyltin (BT) and phenyltin (PT), in aquatic organisms from Merambong and Tinggi Island, Malaysia, which differ in industrial and economic activities, were measured. Tributyltin (TBT) compounds among BTs ranged from 2.9–28 and <0.1–21 ng g−1 ww in aquatic organisms from Merambong and Tinggi Island, respectively. Triphenyltin (TPT) compounds among PTs ranged from <0.1–25 ng g−1 ww and <0.1–61 ng g−1 ww in aquatic organisms from Merambong and Tinggi Island, respectively. A survey of OT concentrations among the species of OT compounds revealed that the concentrations of BTs and PTs in crabs and clams were high while those in fish were low. Correlation analysis with stable nitrogen isotopes suggest that OT compounds did not accumulate in aquatic organisms through the food web. The measurement of OT compounds in tissues and organs of fish revealed that BT concentrations in liver were higher than those in muscle and that BTs were detected in eggs of fish.

To elucidate the details of both the current status of contamination and the accumulation of organotin compounds (OTs) in mangroves in coastal ecosystems, we determined the concentrations of butyltin compounds (BTs) and phenyltin compounds (PTs) in sediment and mangrove leaves collected from mangrove forests in Merambong and Tinggi Island, Malaysia. Butyltins were detected in all sediment samples collected from both regions, whereas PTs were not detected. The levels of tributyltin (TBT) compounds in the sediment samples were lower than those from previous studies conducted in Malaysia and at other sites in South-east Asia. In both Merambong and Tinggi Island, the average proportions of dibutyltin (DBT) and monobutyltin (MBT), which are degradation products of TBT, were ~70%. This result suggests that the input of TBT has decreased in Malaysia. The proportions of DBT and MBT in the sediment from Merambong were higher than those from Tinggi Island. The concentrations of TBT in mangrove leaves from Tinggi Island were significantly higher than those from Merambong. MBT was the most dominant type among the BTs in mangrove leaves from both Merambong and Tinggi Island. The ratios of the BTs burden in mangrove leaves to the BTs concentration in sediment from Merambong and Tinggi Island averaged 3.1 and 6.2, respectively. Among the values of BTs, the MBT values were found to be the highest in both regions.

The susceptibility of marine bacterial communities to copper pyrithione (CuPT2), zinc pyrithione (ZnPT2) and their degradation product is described and toxicities of these relatively new antifouling biocides compared with those of their harmful organotin (OT) predecessors, tributyltin (TBT) and triphenyltin (TPT). These biocides were added to agar at concentrations of 0, 0.01, 0.1, 1 and 10 mg l−1 and coastal seawater including indigenous bacteria added to each batch of agar solution. The number of bacterial colony forming units (CFU) was measured after 7 days culture. Relative CFU (as a percentage of control) was more than 80% at a concentration of 0.01 mg l−1 of each compound, except for TBT. Relative CFU decreased as a function of dose of each biocide, although concentration-dependent changes in rate of CFU were relatively low during exposure to degradation products of CuPT2 and ZnPT2, pyridine N-oxide (PO) and pyridine-2-sulphonic acid (PSA). Based on comparisons of EC50, TBT was the most bacterio-toxic of the tested compounds (0.2 mg l−1), marginally more so than CuPT2 (0.3 mg l−1). Interestingly, EC50 values of degradation products of CuPT2 and ZnPT2, 2-mercaptopyridine N-oxide (HPT) and 2,2′-dithio-bispyridine N-oxide (PT2) were 0.8 and 0.5 mg l−1, respectively, lower than that of the parent chemical, ZnPT2 (1.4 mg l−1). The EC50 of PT2 was also lower than that of TPT (0.7 mg l−1), implying higher toxicity. Given the overlapping toxicity ranges, these results suggest that marine bacterial communities experience comparably high susceptibility to metal PTs and OTs during their life history.

To examine the risk of transgenerational transfer of organotin compounds (OTs) in fish, tributyltin (TBT) and triphenyltin (TPT) compounds and their breakdown products were determined in both parental females and offspring of viviparous surfperch Ditrema temmincki collected from Japanese coastal waters. TBT concentrations (Mean ± SD) in the offspring (34 ± 5.7 ng Sn g−1 wet wt) were significantly higher (10–17 times) than in the parental females (2.8 ± 1.0 ng Sn g−1 wet wt). In the offspring, TBT was the predominant butyltin compound (82 ± 1.6% ∑BTs = TBT + DBT + MBT), and represented a greater proportion than in the parental females (51 ± 9.3% as TBT). TPT concentrations were significantly lower than TBT, and the ratio of TPT in parental females, relative to offspring, was different from TBT. TPT concentrations in the offspring (0.8 ± 0.3 ng Sn g−1 wet wt) were almost identical to those in the parental females (1.0 ± 0.5 ng Sn g−1 wet wt). TPT was the predominant phenyltin (∑PTs = TPT + DPT + MPT) in both offspring (73 ± 12% as TPT) and parental females (72 ± 18% as TPT). Results suggest that the transfer rate of TBT from parent to offspring could be much faster than its degradation rate in the offspring, accounting for higher accumulation of TBT in the latter. In contrast, the transfer rate of TPT is slower than its biodegradation, leading to a lower concentration of TPT in the offspring. It is therefore likely that the offspring might be at a higher risk from TBT than the parental females during their early growth stage in ovary in the viviparous surfperch whereas exposure to TPT is comparable in both generations.

The imposition of male sexual characteristics onto the female (imposex) is present in wild populations of the non-native veined rapa whelk (Rapana venosa) in Chesapeake Bay, USA but does not appear to compromise reproductive function. Cultured whelks were used to test two hypotheses: (1) Observed imposex metrics will be similar to tributyltin (TBT) water concentrations at each of three sites; (2) Male and imposex/female whelks from the same site will have similar TBT body burdens. Cultured 2-year-old whelks were transplanted to three field sites in the York River, USA at the onset of their second reproductive season. Transplant site mean TBT water concentrations ranged from 1.4 ± 0.77 to 64.2 ± 57.8 ng l−1. Imposex incidence was 100% after 28 weeks with an observed M:F:IF ratio of 81:0:92 across all sites. Imposex stages (median vas deferens scale index = 4) and reproductive output were similar across sites. The imposex severity (IS = penis length/shell length) increased with increasing TBT concentrations. The relative penis length (RPLI) and relative penis size (RPSI) indices were positively related to site-specific TBT levels. Male whelks accumulated significantly higher TBT concentrations than female whelks at the site with the highest TBT concentration. Mean TBT concentrations in whelk egg capsules were significantly higher than concentrations in male or female whelk tissue. Egg capsule deposition provides a depuration mechanism for female whelks to reduce body burden of lipophilic TBT. Sex, season and reproductive status should be considered when using gastropod bioaccumulation to monitor TBT effects.

European Community regulations on chemicals promote alternative methods to test substances presenting potential risks for the environment. In the present work, cultured atrial cells isolated from oyster (Crassostrea gigas) were used as an experimental model to investigate the toxicity of tributyltin (TBT) after short-time exposure at concentrations representative of those that can be measured in seawater, marine sediments and/or bivalves bioaccumulating this pollutant. In vitro and in vivo assays produce values of the same order of magnitude for both animal/cell survival and heart/cardiomyocyte beating rate. The survival rate of whole animals decreased from 10-6 M TBT after 3 days. For cultured cells, the viability, evaluated using 3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) assay, significantly decreased after two days of treatment with 10-6 M TBT, and after six days with 10-10 M TBT. The percentage of apoptotic cells, quantified by flow cytometry and YO-PRO®-1 iodide, a nucleic acid stain that only permeates cells that are beginning to undergo apoptosis, increased significantly in these cases. Moreover, intracellular concentration of Ca++ had increased after 10 min of exposition to 10-6 M, and could be associated with apoptotic processes. As patch clamp experiments showed that Ca++ conductance was decreased, intracellular calcium increase could mainly be due to a release from internal stores. The decreases in beating rhythm could be explained by the decrease in adenosine triphosphate (ATP) production revealed by 31 P nuclear magnetic resonance (NMR) spectroscopy and confirmed by the increase of the KATP channel conductance. The related hyperpolarization and the disturbances of the energetic metabolism were clearly related to the loss of the atrial cell contractility and viability.

The dogwhelk Nucella lapillus experienced localized extinction in the 1980s and 1990s due to the use of tributyltin (TBT) antifoulants, causing imposex in females. The aim of this study was to establish the extent of the return of the species across the mainland coast of central southern England as TBT use has been progressively restricted, and to quantify the extent of imposex impact on the populations present. We surveyed from Poole to Selsey where isolated populations had become extinct, and the Isle of Wight where some populations had persisted. We found evidence that since TBT restrictions, recolonization and colonization by N. lapillus has been rapid. By 2007–2008, of the eleven surveyed mainland sites, seven were colonized, although indications of reduced imposex impacts were mixed. Distribution had also extended on the Isle of Wight and populations were larger with less imposex impact in sites with long term populations. The lack of continuous suitable habitat blocks and the hydrodynamic complexity of the region, leads us to hypothesize that recovery has been facilitated by man-made structures which may be acting as ‘stepping stones’. Populations that have become established on engineered structures such as sea walls, breakwaters and rock groynes demonstrate accelerated recovery in the region as TBT in the environment has generally declined. Sites with suitable substrates and food sources near to ports were either not recolonized or had small populations with imposex evident. For species with a short pelagic larval stage or with direct development, population connectivity between patches of harder substrata along hydrodynamically complex coastlines may be greater than previously thought.

Most neogastropod species exhibit masculinization of the female when subject to tributyltin (TBT) pollution (a process known as ‘imposex’). To date, the dog-whelk Nucella lapillus is seemingly unique in having a genetic deficiency (termed Dumpton syndrome or DS) that disrupts the development of normal male sex organs, its presence being readily recognizable by the underdevelopment, or non-development (aphally), of the penis, and incomplete formation (non-closure) of the vas deferens, causing a split prostate. In highly contaminated conditions, female carriers of DS can be identified by a lesser degree of masculinization (notably aphally): they escape sterilization caused by the advanced stages of imposex. To date, DS has only been reported in areas with high TBT pollution which induces sterilization of normal females (i.e. non DS-affected females). DS is now, for the first time, observed at some locations where present TBT levels are low and some normal females lack penis development. In such conditions it is not possible to discriminate normal from DS-affected females using aphally. As DS-affected females must be discarded from the calculation of the imposex bioindicators to monitor TBT pollution, indirect tools such as molecular probe are now needed to further survey those areas where DS and TBT pollution may interact as, for example, in south-west Brittany.

Shell malformations and histological effects of environmental concentrations of TBTF (2, 13.1 and 64.8 ng/l) were assessed on adult oysters Crassostrea gigas during a 1-month experiment, including both an exposure and a depuration phase. The results showed that the digestive gland is the primary target organ. Recoverable modifications were observed at the lowest concentration. Limited necrosis was observed at the highest concentration and a longer exposure could have led to extensive and irreversible tissue lesions. Shell malformations (chambering) were observed during the depuration phase. Finally the authors suggest that safe TBT levels in mariculture waters should be lower than 2 ng/1.
