DNA polymerases play a central role in biology by transferring genetic information from one generation to the next during cell division. Harnessing the power of these enzymes in the laboratory has fueled an increase in biomedical applications that involve the synthesis, amplification, and sequencing of DNA. However, the high substrate specificity exhibited by most naturally occurring DNA polymerases often precludes their use in practical applications that require modified substrates. Moving beyond natural genetic polymers requires sophisticated enzyme-engineering technologies that can be used to direct the evolution of engineered polymerases that function with tailor-made activities. Such efforts are expected to uniquely drive emerging applications in synthetic biology by enabling the synthesis, replication, and evolution of synthetic genetic polymers with new physicochemical properties.

The new generation of energy-dispersive X-ray (EDX) detectors with higher count rates than ever before, paves the way for a new approach to quantitative elemental analysis in the scanning transmission electron microscope. Here we demonstrate a method of calculating partial cross sections for use in quantifying EDX data, beneficial especially because of the simplicity of its implementation. Applying this approach to acid-leached PtCo catalyst nanoparticles leads to quantitative determination of the Pt surface enrichment.

We report the synthesis of carbon nanowires (CNWs) via chemical vapor deposition using catalytic decomposition of ethanol on nanosized transition metals such as Co, Fe, and Ni. Dip-coating process was used for the formation of catalytic nanoparticles, inducing the growth of CNWs on the surface of the carbon fiber paper (CFP). The liquid ethanol used as carbon source was atomized by an ultrasonic atomizer and subsequently flowed into the reactor that was heated up to a synthesis temperature of 600–700°C. Microscopic images show that CNWs of <50 nm were densely synthesized on the surface of the CFP. Raman spectra reveal that a higher synthesis temperature leads to the growth of higher crystalline CNWs. In addition, we demonstrate the successful decoration of platinum nanoparticles on the surface of the prepared CNWs/CFP using the electrochemical deposition technique.

Equations governing the elastic scattering of electrons are applied to the knock-on displacement of atoms along a substrate, yielding analytical expressions for the surface-translation energy, threshold incident energy, and displacement rate. For a surface perpendicular to the incident beam, scattering angles around 90° contribute most to the kinetic energy of surface atoms. Tilting the specimen lowers the threshold incident energy for displacement and leads to anisotropy in the atomic motion but has little effect on the directionally-averaged displacement rate. The rate of beam-induced adatom motion is predicted to exceed that of room-temperature thermal motion when the surface-diffusion energy is greater than about 0.5 eV.

In prior research, specimen holders that employ a novel MEMS-based heating technology (AduroTM) provided by Protochips Inc. (Raleigh, NC, USA) have been shown to permit sub-Ångström imaging at elevated temperatures up to 1,000°C during in situ heating experiments in modern aberration-corrected electron microscopes. The Aduro heating devices permit precise control of temperature and have the unique feature of providing both heating and cooling rates of 106°C/s. In the present work, we describe the recent development of a new specimen holder that incorporates the Aduro heating device into a “closed-cell” configuration, designed to function within the narrow (2 mm) objective lens pole piece gap of an aberration-corrected JEOL 2200FS STEM/TEM, and capable of exposing specimens to gases at pressures up to 1 atm. We show the early results of tests of this specimen holder demonstrating imaging at elevated temperatures and at pressures up to a full atmosphere, while retaining the atomic resolution performance of the microscope in high-angle annular dark-field and bright-field imaging modes.

Gold nanoparticles supported on titania catalysts with different Au loadings were prepared and evaluated in the reaction of NO reduction by CO in an oxygen rich condition. The crystalline structures of the Au/TiO2 materials were refined with Rietveld method. TiO2 support chiefly contains anatase phase, having a crystalline size ranged from 5 to 15 nm. Au particles have an average crystal size approximately 2-5 nm as Au concentration less 3 wt %. In the reaction of NO + CO + O2, the Au/TiO2catalysts show a selectivity to 100 % N2, neither NO2 nor N2O was yielded in the reaction temperature between 25 and 400 °C, which strongly indicates that Au/TiO2catalysts are much superior to the other catalysts like Pt/TiO2 catalysts on which N2O was usually produced in the reaction temperature below 200 °C and NO2 was produced in the reaction temperature above 300 °C under a similar reaction condition.

Ordered nanoscale pore systems such as those represented by zeolites offer many opportunities for the design of complex functional systems via self-assembly.With their large internal surface areas and tunable, well-defined crystalline pore structures that allow molecular sieving and ion exchange, zeolites can be adapted for numerous applications. The nanoscale reactors present in zeolite pore systems have been explored as structural templates for the spatial organization of numerous guests. Examples from various fields are discussed, such as the stabilization of organic dyes for the construction of energy transfer and storage systems, the construction of host–guest hybrid catalyst systems, and the encapsulation of conducting or semiconducting nanoscale wires and clusters. More complex, hierarchical forms of nanostructured matter become accessible when zeolite crystals are used as building blocks for the selfassembly of thin films or three-dimensional objects. A combination of weaker and stronger interactions ranging from dispersive forces, hydrogen bonding, and electrostatic interactions to covalent bonding can be used to build functional hierarchical constructs. Several examples and novel applications of such systems will be discussed, including oriented channel systems, chemical sensors, and hierarchical pore systems for catalytic reactions.

The structural and chemical heterogeneity of 2.5-nm Pt50Ru50 electrocatalysts was studied by transmission electron microscopy using selected area diffraction, lattice imaging, electron-energy loss spectroscopy, and energy-dispersive X-ray spectroscopy. The catalysts with the highest methanol oxidation activities exhibit oxidation-induced phase separation on the nanoscale to form Pt-rich metal embedded in Ru-rich hydrous and anhydrous oxide. Reduction of the oxide-on-metal samples produces a true bimetallic face-centered cubic Pt50Ru50 alloy, with 275 times lower oxidation activity.
