Members of the genus Drepanocephalus are endoparasites of fish-eating birds of the families Phalacrocoracidae and Sulidae distributed across the Americas. Currently, Drepanocephalus contains three species, i.e. D. spathans (type species), D. olivaceus and D. auritus. Two additional species, D. parvicephalus and D. mexicanus were transferred to the genus Petasiger. In the current study, available DNA sequences of D. spathans, D. auritus and Drepanocephalus sp., were aligned with newly generated sequences of D. spathans and Petasiger mexicanus. Phylogenetic analyses inferred with three nuclear (LSU, SSU and ITS1, 5.8S, ITS2) and two mitochondrial (cox1, nad1) molecular markers showed that the sequences of D. spathans and D. auritus are nested together in a single clade with very low genetic divergence, with Petasiger mexicanus as its sister species. Additionally, P. mexicanus was not a close relative of other members of the genus Petasiger, showing that P. mexicanus actually belongs to the genus Drepanocephalus, suggesting the need to re-allocate Petasiger mexicanus back into the genus Drepanocephalus, as D. mexicanus. Morphological observations of the newly sampled individuals of D. spathans showed that the position of the testes is variable and testes might be contiguous or widely separated, which is one of the main diagnostic traits for D. auritus. Our results suggest that D. auritus might be considered a synonym of D. spathans and, as a result, the latter represents a species with a wide geographic range across the Americas, parasitizing both the Neotropical and the double-crested cormorant in Argentina, Brazil, Paraguay, Venezuela, Colombia, Mexico, USA and Canada.

Despite the fact that tapeworms comprise the bulk of parasite communities of sharks in marine ecosystems, little is known about their life cycles and, more specifically, about the potential intermediate hosts they utilize as transmission routes. In the absence of morphological features required for specific identification of larval tapeworms from potential intermediate hosts, recent molecular advances have contributed to linking larval and adult parasites and, in some instances, uncovering unknown trophic links. Host–parasite checklists are often the first source of information consulted to assess the diversity and host specificity of parasites, and provide insights into parasite identification. However, these host–parasite checklists are only useful if they encompass the full spectrum of associations between hosts and parasites. A checklist of New Zealand fishes and their parasites has been published, but recent parasitological examinations of commercial fish species reveal that the checklist appears to be far from complete. We focused our current study on a comprehensive survey of macroparasites of a commercial species, the New Zealand sole (Peltorhamphus novaezeelandiae) off the coast of Otago, New Zealand. Specifically, we were expecting to recover marine tapeworms using sharks as their definitive hosts that are generally underreported in parasite surveys. The parasites recovered included tapeworms, flukes, round worms and thorny-headed worms. Surprisingly, a large proportion of the non-tapeworm parasites we recovered were not previously reported from this fish species. A discussion on the potential ecological roles played by this fish species in the transmission of parasites is included.

The cestode Hymenolepis microps is an intestinal parasite of tetraonid birds, including the willow ptarmigan (Lagopus lagopus). This parasite is able to maintain a high prevalence and intensity throughout the year, even in a subarctic environment in bird populations with relatively low host densities, indicating effective transmission routes. Willow ptarmigan consume mainly vegetal material and active consumption of invertebrates is confined to the first two or three weeks of life. Ptarmigan are infected by different species of ectoparasites, of which two species of feather lice, Lagopoecus affinis and Goniodes lagopi, are the most abundant. In this study, we explored the hypothesis that feather lice may be suitable intermediate hosts for H. microps. We applied histological techniques and light microscopy to investigate lice for the presence of larval cestode stages (cysticercoids). We found 12 cysticercoid-like structures inside chewing lice collected on L. lagopus hosts harbouring H. microps. In addition, a polymerase chain reaction (PCR) screening of Ischnocera lice DNA, targeting the 18S rRNA gene of the cestode, showed positive results for two different short fragments of the 18S rRNA gene of H. microps which were sequenced from lice collected on birds. Both independent lines of evidence support the hypothesis that Ischnocera lice might be suitable intermediate hosts in the life cycle of H. microps in L. lagopus.

Timoniella spp. are cryptogonimid flukes (Digenea: Cryptogonimidae) that parasitize the guts of fish in brackish waters. Timoniella imbutiforme, a species from the Mediterranean Sea, is recorded in the Black Sea, while T. balthica has been described from the Baltic Sea. In this paper, we clarify the taxonomic status of Timoniella populations in the Baltic and Black seas. Adults and metacercariae of Timoniella spp. were sampled from localities in the Mediterranean Sea (France), Black Sea (Ukraine) and Baltic Sea (Germany) and subjected to molecular and morphological analysis, including Bayesian phylogenetic reconstruction based on concatenated sequences of ITS1–ITS2–28S. This allowed us to construct a new key to species of the genus Timoniella. Our results suggest that T. balthica forms part of the Boreal–Atlantic relict fauna of the Black Sea and should now be considered a junior synonym of T. imbutiforme.

Cryptic parasite diversity is a major issue for taxonomy and systematics, and for attempts to control diseases of humans, domestic animals and wildlife. Here, we re-examine an earlier report that, after correcting for sampling effort, more cryptic species of trematodes are found per published study than for other helminth taxa. We performed a meta-analysis of 110 studies that used DNA sequences to search for cryptic species in parasitic helminth taxa. After correcting for study effort and accounting for the biogeographical region of origins, we found that more cryptic species tend to be uncovered among trematodes, and fewer among cestodes and animal-parasitic nematodes, than in other helminth groups. However, this pattern was only apparent when we included only studies using nuclear markers in the analysis; it was not seen in a separate analysis based only on mitochondrial markers. We propose that the greater occurrence of cryptic diversity among trematodes may be due to some of their unique features, such as their mode of reproduction or frequent lack of hard morphological structures, or to the way in which trematode species are described. Whatever the reason, the high frequency of cryptic species among trematodes has huge implications for estimates of parasite diversity and for future taxonomic research.

Two new and one known species of Annulotrema Paperna & Thurston, 1969 are reported from the gills of the tigerfish Hydrocynus vittatus Castelnau, 1861, collected in Lake Kariba, Zimbabwe. The new species, Annulotrema pseudonili n. sp. and A. bracteatum n. sp., are described and distinguished mainly on the basis of features of the male copulatory organ (MCO). Annulotrema pseudonili n. sp. most closely resembles A. nili Paperna, 1973, but differs from it by possessing a more delicate MCO with a thin-walled base without a fibrous distal part. Annulotrema bracteatum n. sp. is most similar to Annulotrema ruahae Paperna, 1973, from which it differs by having an MCO composed of a longer copulatory tube and a leaf-shaped accessory piece enveloping the distal part of the tube. The presence of Annulotrema pikoides Guégan, Lambert & Birgi, 1988 on H. vittatus in Zimbabwe represents a new locality record for this parasite.

Increasing urbanization along riverbanks is a constant source of stressors to the aquatic community, and the use of bioindicators is suitable to detect and monitor the effect of each stressor. We investigated the parasites of the ‘cará’ fish (Geophagus brasiliensis) as potential bioindicators in a river whose banks are subject to increasing anthropogenic pressure. Samples were taken at four points of the Sangradouro River, in Florianópolis, southern Brazil, bimonthly for 12 months. Water temperature, pH, dissolved oxygen, conductivity and salinity were measured at each point and water samples were taken for nutrient analysis (total nitrogen, nitrite, total ammonia nitrogen, total phosphorus, phosphate), and total and faecal coliforms. A generalized linear model (GLM) was constructed using the abundance of each parasite species as the response variable and biometric characteristic of the fish and water variables as possible predictors. Among the 137 fish examined, 114 (83.2%) were parasitized by at least one parasite species. Two species of ectoparasites (Sciadicleithrum guanduensis and Neascus-type metacercariae) and two species of endoparasites (Pandosentis aff. iracundus and Homalometron pseudopallidum) were observed. This is the first record of the genus Pandosentis in Brazilian waters and the first record of the species G. brasiliensis as a host for this parasite. Among the analysed groups of parasites, monogeneans and acanthocephalans proved to be the most sensitive to the concentrations of dissolved oxygen, faecal coliforms and total ammonia nitrogen in the water. Our study suggests that the abundance of both monoxenic and heteroxenous parasites can be negatively affected by organic pollution in the aquatic environment, reinforcing the potential of fish parasites as indicators of water quality.

Cystic echinococcosis (CE) is a medically important parasite-caused human disease. Humans may acquire the infection accidentally by ingestion of E. granulosus eggs. The parasite has a broad range of hosts and genotypes, which may affect its aetiological and biological characteristics. The present study aimed to determine the genetic characteristics of human isolates of E. granulosus in Isfahan, Iran. In this cross-sectional study, 50 surgically removed hydatid cysts were collected from hospitalized patients in Al-Zahra Hospital, Isfahan, Iran, over a period of 2 years (2015–2017). DNA was extracted from cyst material, and polymerase chain reactions (PCR) were performed targeting cox1 and nad1 genes. Amplicons were sequenced directly and the resulting sequences were aligned and analysed. Phylogenetic and genetic diversity analyses were also performed. Among the isolates, 43 (86%), 3 (6%) and 4 (8%) out of 50 were E. granulosus (G1), E. granulosus (G3) and E. intermedius (G6), respectively. In total, nine and eight haplotypes were identified by nad1 and cox1 gene analysis, respectively. The haplotype diversity index was higher by cox1 gene analysis (0.547) in G1 strains compared with nad1 (0.433). The G1 genotype was the most predominant isolate from human cases of CE, and the presence of G6 is indicative of an important role of camels in the development of human CE in Isfahan. This is the first report of the G3 genotype causing human CE in Isfahan. Moreover, cox1 gene analysis enables a higher resolution of the genetic diversity of the E. granulosus population compared with nad1 gene analysis.

Infection by Fasciola species was investigated in seven districts of Dakhla Oasis, Egypt, through abattoir inspection of cattle livers for adult worms and sedimentation of faecal samples from local cattle to detect Fasciola eggs. In addition, lymnaeid snails collected from the study area were examined microscopically for developmental stages of Fasciola spp. Abattoir inspection revealed that 51 out of 458 cattle livers (11.1%) contained adult flukes, which were identified morphologically as Fasciola hepatica. Examination of the cattle faecal samples revealed that 142 out of 503 (28.2%) contained Fasciola eggs. The collected snails, identified as Galba truncatula and Radix natalensis, showed larval stages of Fasciola in 71 out of 731 (9.7%) G. truncatula, while R. natalensis showed no infection. Specific duplex polymerase chain reaction (PCR) targeting the mitochondrial cox1 gene of F. hepatica and Fasciola gigantica was carried out on DNA extracted from pooled infected snails and adult worms. The F. hepatica size amplicon (1031 bp) was obtained from both the infected G. truncatula and the adult worms isolated from cattle livers from different districts. The amplicon sequences were identical to the published sequences of F. hepatica mitochondrial cox1 gene. In conclusion, the zoonotic importance of Fasciola infection and appropriate hygienic measures must be taken into consideration in Dakhla Oasis, Egypt.

Species-specific microenvironmental preferences and interactions between parasite species have been the focus of many ecological studies. Here, we studied the distribution of ectoparasite species within the gill apparatus of bream (Abramis brama) from Lake Lubāns (Latvia) to establish whether digenean metacercariae: (1) prefer specific patches within the gill apparatus; (2) co-occur in the same patches with monogeneans and copepods within a host individual; and (3) interact with monogeneans and copepods. We recorded all parasites on gill arches of the same host species and used null models to analyse co-occurrences of digenean metacercariae, monogeneans and copepods. Zero-inflated mixture models were used to define the preferred patches of parasites. We found that digenean metacercariae (Bucephalus polymorphus) prefer specific patches of the gill apparatus to encyst, and shared these preferences with monogeneans and copepods, but did not interact with them. We concluded that digenean metacercariae have a species-specific microenvironmental preference to encyst in the gill apparatus and their occurrence (even in high numbers) does not reduce the success of attachment of monogeneans and copepods in the same gill patches.

The complex life cycle of taeniids represents an ideal model of a multi-host system. The complexity of these parasites can therefore cover the epidemiological issues of the interface between wild and domestic animals, especially once spatial overlap between wild and domestic definitive and intermediate hosts occurs. Here we use the occurrence of Taenia ovis krabbei in two model areas as an example of this epidemiological complexity. In two contiguous areas in the Italian northern Apennines, two hunted roe deer (Capreolus capreolus) showed numerous cysticerci in the muscles of their whole body and an adult tapeworm was recorded in a semi-stray dog (Canis lupus familiaris). Through molecular typing of the mitochondrial cytochrome c oxidase I (cox1) gene, cysticerci and the adult tapeworm of T. krabbei were identified. Taenia krabbei cysticercosis was recorded for the first time in Italy. Although the role of dogs in the parasite's life cycle emerges, the overlap between wild and domestic definitive hosts and the increase of wild population densities raise concerns about the temporal (old or new) introduction and the spread of this parasite by one of these canid species (wolf (Canis lupus) or dog). Although T. krabbei is not a public health issue, economic concerns emerged for hunters and meat producers, related to the damage of carcasses by cysticerci. Therefore, there is a need to evaluate the spread of T. krabbei in the intermediate and definitive host populations, and to ensure the relevant sanitary education for hunters in order to avoid practices that could favour the spread and maintenance of its life cycle.

As a result of experimental studies conducted in the Russian southern Far East, adult worms from the genus Metagonimus were obtained. A comparative analysis of the morphometry of these worms with other Metagonimus representatives showed that they are most similar to M. katsuradai Izumi, 1935 and M. otsurui Shimazu & Urabe, 2002 found in Japan, due to the ratio of suckers and the positions of the testicle, uterus and vitellaria. However, Russian worms differ from species in Japan by other metric characters: they differ from M. otsurui by the maximum size of most organs and from M. katsuradai by body width, pharynx length, and maximum size of testes and ovary. At the same time, they are identical to a trematode from the Russian southern Far East, which was previously identified as M. katsuradai. The validity of this species was also confirmed by genetic data. According to the 28S gene and the internal transcribed spacer 2 (ITS2) region of rDNA, as well as the cytochrome c oxidase I (cox1) gene of mtDNA, the Metagonimus specimens found in Russia differ from published genetic data for other members of this genus. However, both morphological similarity and molecular data showed that M. pusillus sp. nov., M. katsuradai and M. otsurui are most likely cryptic species. Furthermore, additional data based on a mitochondrial marker were provided for M. suifunensis Shumenko, Tatonova & Besprozvannykh, 2017 from Russia.

We describe Isorchis cannoni n. sp. from the rabbitfishes Siganus fuscescens (Houttuyn) and Siganus lineatus (Valenciennes) (Siganidae) collected off Heron Island, southern Great Barrier Reef, Australia and, using molecular data, demonstrate that ‘Cercariae queenslandae II’ of Cannon (1978) from the gastropod Clypeomorus batillariaeformis Habe & Kosuge (Cerithiidae) is the larval form of this new species. The cercariae of I. cannoni n. sp. develop in rediae, encyst in the environment after emergence, and are inferred to then be consumed by grazing rabbitfish. Additionally, we provide a new report of Isorchis currani Andres, Pulis & Overstreet, 2016 from the type host, Selenotoca multifasciata (Richardson) (Scatophagidae) collected in Moreton Bay, south-east Queensland, Australia, greatly expanding the known geographical range of this species. Molecular sequence data (ITS1, ITS2 and 28S rDNA) generated for I. cannoni n. sp. and the new specimens of I. currani, confirm the identification of I. currani and demonstrate a distinct genotype for I. cannoni n. sp. relative to other species of Isorchis Durio & Manter, 1969, for which molecular data are available. Isorchis cannoni n. sp. is morphologically distinct from all other species in the genus, and is further distinguished by utilizing species of Siganidae as definitive hosts, rather than species of Chanidae or Scatophagidae. Because haploporid and atractotrematid cercariae have well-developed reproductive organs, we find cercariae of these closely related families morphologically distinguishable in the same way as adult trematodes: atractotrematids have two symmetrical testes and haploporids have a single testis or, rarely, two tandem or oblique testes.

The musk deer (Moschus berezovskii) is an economically important species from which musk is extracted and used in perfumes and medicines. Cestodes (parasitic flatworms) of the genus Moniezia are important parasites that infect this endangered species and can cause high mortality in young deer. In 1982, Moniezia (S.) sichuanensis sp. nov. was described from a specimen obtained from wild musk deer. The new species was distinct from the other described species of Moniezia by the sawtooth-shaped interproglottidal glands, the thick vagina and the absence of a cirrus spine. In the present study, 12 cestodes collected from musk deer were examined morphologically and confirmed to be M. sichuanensis. Molecular characterization was performed by amplifying and comparing the internal transcribed spacer 1 (ITS1) and 5.8S rRNA gene (ITS1–5.8S) of ribosomal DNA with available sequences from other Moniezia species. The amplified sequences ranged from 761 to 764 bp and similarity ranged from 98.7–100%, compared to 67.8–92.4% with other Moniezia spp. Construction of a phylogenetic tree using the neighbour-joining method indicated that all 12 ITS1–5.8S sequences formed a single clade, confirming M. sichuanensis as a separate species. This study provides novel molecular insight into M. sichuanensis that could prove useful for future diagnosis and control of monieziasis in musk deer.

Environmental changes and ecological disturbances can have large and unpredictable effects on parasite dynamics. Increasing human impacts on freshwater ecosystems through land use may thus modify the distribution and abundance of parasites and have cascading effects on host populations. Here we tested the effects of small-scale riparian forest management on the nematode Cystidicoloides ephemeridarum and its insect intermediate host Ephemera danica in forested streams. We assessed the impacts of harvesting riparian trees on parasite prevalence and abundance concomitantly with host densities. We also looked at upstream and downstream reaches to document potential cascading effects on unaltered stream sections mediated by aerial dispersal of adult mayfly or downstream drift of E. danica larvae. We show that host densities and parasite levels (prevalence and abundance) increased significantly following riparian tree removal. Overall, parasite densities showed a 6- to 66-fold increase in harvested reaches compared to upstream, pristine reaches. Similar effects were also clear downstream of the disturbance. Thus, despite the small extent of riparian forest alteration along the study streams, both parasite and intermediate host were strongly affected. Small-scale riparian forest management may thus have large, unforeseen impacts on some aspects of freshwater ecosystem structure and functioning that are often ignored. Generally, understanding how human perturbations influence parasites is vital when trying to predict overall impacts on ecosystem structure and functioning, and how changes in infection dynamics may further affect host species.

The aims of the present study were to identify and describe the occurrence of nematode parasites in the gonads of bluefish Pomatomus saltatrix from off the coast of the state of Rio de Janeiro, Brazil. Only females were found to be parasitized by the nematodes, which were identified as P. saltatrix using morphological, morphometric and molecular data. Infection of female bluefish by this nematode had the following values: prevalence, 48.7%; mean intensity, 2.6; mean abundance, 1.3; and range of infection, 1–10 specimens. Histopathological examination of transverse and longitudinal sections of the parasitized ovaries showed nematodes at different stages of development among oocytes, but no indication of any associated inflammatory reaction. The presence of nematodes in the ovaries of bluefish is an important indication of fish hygiene, and parasitized fish are usually rejected by consumers because of their repugnant appearance.

A new species of cestode of the genus Fuhrmannetta found in the small intestine of Cerradomys goytaca is described herein, named Fuhrmannetta jurubatensis n. sp. Rodents were collected from the sand-plains areas of the northern coast of the State of Rio de Janeiro, Brazil. Morphological analyses were conducted by light and scanning electron microscopy. From the morphological analysis and a comparison with the known species of the genus, F. jurubatensis n. sp. can be identified by a combination of morphological and morphometrical characteristics, including strobila length, number and length of rostellar hooks, position of the genital pore and the number of eggs per uterine capsule.

In the present study, phylogenetic relationships of European and Far Eastern representatives of the genus Aspidogaster Baer, 1827 were analysed: A. conchicola Baer, 1827, A. limacoides Diesing, 1834, A. ijimai Kawamura, 1915 and A. chongqingensis Wei, Huang & Dai, 2001. Based on ITS1–5.8S–ITS2 rDNA sequence data, an obvious differentiation was seen between specimens of A. limacoides sensu stricto from the European part of Russia and A. limacoides sensu Chen et al., 2010 from China (13.7%); the latter parasites were recognized as A. chongqingensis. Aspidogaster chongqingensis was more closely related to A. ijimai than to A. limacoides s. str. Specimens of A. ijimai from the Amur River, Khanka Lake (Russian Far East) and China were grouped into a single clade with low intra specific molecular differentiation (d = 0–0.3%). Specimens of A. conchicola from the European part of Russia, the Russian Far East and China also formed a single distinct clade. Genetic differentiation between European and Chinese samples of this species was two times lower (d = 0.45%) than between Russian Far East and European or Chinese samples (d = 0.96%), suggesting a long-term separate existence of A. conchicola in the Russian Far East.

Eurytrema pancreaticum is one of the most common trematodes of cattle and sheep, and also infects humans occasionally, causing great economic losses and medical costs. In this study, the sequences of the complete nuclear ribosomal DNA (rDNA) repeat units of five E. pancreaticum individuals were determined for the first time. They were 8306–8310 bp in length, including the small subunit (18S) rDNA, internal transcribed spacer 1 (ITS1), 5.8S rDNA, internal transcribed spacer 2 (ITS2), large subunit (28S) rDNA and intergenic spacer (IGS). There were no length variations in any of the investigated 18S (1996 bp), ITS1 (1103 bp), 5.8S (160 bp), ITS2 (231 bp) or 28S (3669 bp) rDNA sequences, whereas the IGS rDNA sequences of E. pancreaticum had a 4-bp length variation, ranging from 1147 to 1151 bp. The intraspecific variations within E. pancreaticum were 0–0.2% for 18S rDNA, 0–0.5% for ITS1, 0% for 5.8S rDNA and ITS2, 0–0.2% for 28S rDNA and 2.9–20.2% for IGS. There were nine types of repeat sequences in ITS1, two types in 28S rDNA, but none in IGS. A phylogenetic analysis based on the 18S rDNA sequences classified E. pancreaticum in the family Dicrocoeliidae of Plagiorchiata, closely related to the suborder Opisthorchiata. These results provide useful information for the further study of Dicrocoeliidae trematodes.

Adults of Skrjabinolecithum spinosum n. sp. were discovered in Mugil cephalus from the Gulf of Peter the Great in southern Far-East Russia. Additionally, adults of Unisaccus tonkini n. sp. were found in the intestine of Moolgarda cunnesius and Moolgarda seheli from the coastal waters of Cat Ba Island, Tonkin Bay, northern Vietnam. Skrjabinolecithum spinosum n. sp. possesses a larger body, and ventral and oral sucker size in comparison with Skrjabinolecithum vitellosum, a smaller pharynx size and body length/width rate ratio in comparison to Skrjabinolecithum pyriforme, a smaller body length and prepharynx size in comparison to Skrjabinolecithum lobolecitum and a smaller pharynx length and egg size in comparison to Skrjabinolecithum indicum and S. lobolecitum. The new species also differs from S. indicum, S. lobolecitum and S. vitellosum by the form of the testis, and from the last two species by the presence of a two-branched intestine. The morphometric parameters of S. spinosum n. sp. are similar to those of Skrjabinolecithum spasskii. However, S. spinosum n. sp., unlike S. spasskii, has an armed hermaphroditic duct. Unisaccus tonkini n. sp. is similar to Unisaccus spinosus (Martin, 1973), Unisaccus brisbanensis (Martin, 1973) and Unisaccus overstreeti (Ahmad, 1987) in body size but differs in oral sucker, pharynx and hermaphroditic sac size from U. spinosus, and in ventral sucker and ovary size from U. brisbanensis and U. overstreeti. Bayesian phylogenetic analysis, based on combined data of internal transcribed spacer 2 (ITS2) and partial 28S rRNA gene sequences, confirmed the validity of S. spinosum n. sp. and U. tonkini n. sp. Analysis of interrelationships of the family Haploporidae, including molecular data on new species, showed that the Waretrematinae subfamily is more heterogeneous in comparison with Haploporinae and Forticulcitinae, and includes U. tonkini n. sp.
