Ulva armoricana sp. nov. is described from the coast of Brittany, France, using the taxonomic criteria established by Bliding, Koeman and van den Hoek for the European species of Ulva. Morphological evidence strongly suggests that Ulva armoricana is conspecific with the free-living Ulva populations responsible for the ‘green tide’ phenomenon that occurs at some eutrophicated sites in North Brittany. Blades of U. armoricana are variable in shape, fragile, easily torn, and 30–55 μm thick in middle and apical regions; they differ from those of U. olivascens by their pale green colour and from those of Ulva lactuca, U. rotundata, U. gigantea, U. pseudocurvata and U. curvata by their crumpled texture that creates an irregular light-reflecting surface, and by the presence of tooth-like protuberances at the margin of the blade. U. armoricana differs from U. scandinavica in having longitudinal ribs containing closely packed bundles of rhizoids, except in the youngest thalli. U. armoricana can be discriminated from U. rigida by the polygonal (never rounded) cells in the mid-thallus region, and quadrangular cells arranged in small longitudinal rows in rapidly growing parts. Pyrenoid number, 1–2 (3–4), overlaps with that of other species, and shape of non-rhizoidal cells of the basal region in transverse sections, either oval or with tapered ends, appears not to be a good taxonomic character. Herbarium data indicate that the species has been collected in northern France since at least 1869, but was previously unrecognized.

The influence of temperature and light on growth and photosynthetic physiology were investigated in embryos of Fucus evanescens grown at 5 or 20°C under irradiances of 15 or 150 μmol photons m−2 s−1 for 7–10 days. Growth was light-independent, but high-temperature embryos were always significantly larger than those grown at low temperature. Photosynthesis-irradiance responses were measured at growth temperature and a standard temperature (20°C) to isolate instantaneous effects of temperature from acclimation responses. Our data indicate that growth and photosynthesis are uncoupled during the early development of Fucus, and that acclimation of the photosynthetic light-harvesting apparatus occurred. Light-limited net photosynthesis (Psub-sat) responded similarly to high temperature and low light. Rates of Psub-sat were similar in embryos grown at 20°C (regardless of light) and at 5°C in low (c. 1.2 nmol O2 mm−3 min−1), whereas those of 5°C high-light embryos were lower (c. −0.04 nmol O2 mm−3 min−1). Changes in Psub-sat were associated with changes in initial slope of the photosynthesis-irradiance curve (α) and dark respiration. Differences in α were attributed to increased absorption due to increased chlorophyll a content and PSII reaction centre densities. Changes in α were also correlated with changes in fluorescence induction kinetics, with high-temperature and/or low-light embryos exhibiting higher ratios of variable: maximum fluorescence (Fv/Fm) than 5°C high-light embryos (c. 0.5 vs. 0.19). In contrast to Psub-sat, changes in light-saturated photosynthesis (Pmax) in response to growth under different temperature/light regimes did not confer metabolic compensation. Rates of Pmax were highest in 20°C high-light embryos (7.3 nmol O2 mm−3 min−1), lower in 20°C low-light and 5°C low-light embryos (c. 2.6 nmol O2 mm−3 min−1) and lowest in 5°C high-light embryos (2.3 nmol O2 mm−3 min−1). We suggest that the ability to achieve temperature-independent rates of Psub-sat may be important for fucoid embryos that recruit in intertidal microhabitats where photosynthesis is often light-limited.

The plastid organelle of plants, eukaryotic algae and apicomplexan protists contains its own DNA genome (the ‘plastome’). In higher plants, at least two RNA polymerases have been shown to participate in the transcription of plastid genes. The multisubunit ‘plastid-encoded plastid RNA polymerase’ (PEP) is derived from the cyanobacterial ancestor of the organelle and is structurally similar to eubacterial RNA polymerases. The core subunits of PEP are encoded by genes on the plastome, whilst sigma-like transcription factors and other regulatory factors are encoded in the nucleus. In contrast, the ‘nuclear-encoded plastid RNA polymerase’ (NEP) is a single-subunit enzyme related to the T3/T7 bacteriophage and mitochondrial RNA polymerases that is believed to have arisen by duplication of the nuclear gene for the mitochondrial enzyme. The situation in algal plastids is less well understood. All algal groups, apart from possibly the dinoflagellates, possess a PEP but it is far from clear whether any possess additional plastid RNA polymerases. In this article we review what is known about the transcription apparatus of algal plastids, and examine the evidence for and against the presence of NEPs.

The taxonomy of Vitreochlamys species (Volvocales, Chlorophyta) was studied with light and electron microscopy; DNA sequence data were obtained for nine strains from various localities in Japan, the Czech Republic, Slovak Republic and Germany. The strains could be delineated clearly into six species – V. fluviatilis (Stein) Batko, V. gloeocystiformis (Dill) Nakazawa, comb. nov., V. ordinata (Skuja) Nakazawa, comb. nov., V. nekrassovii (Korshikov) Nakazawa, comb. nov., V. aulata (Pascher) Batko, and V. pinguis Nakazawa, sp. nov. – on the basis of differences in cell and protoplast shape, numbers of contractile vacuoles and pyrenoids, form and position of the stigma, location of the nucleus, degree of longitudinal striations on the chloroplast surface, and ultrastructure of the pyrenoid and stigma. Although V. pinguis and V. aulata share common chloroplast morphology, including stigma and pyrenoid ultrastructure, these two species can be distinguished clearly by cell shape, the number of contractile vacuoles and location of the nucleus. In addition, phylogenetic sequence data of the rbcL (large subunit of Rubisco, ribulose-1,5-bisphosphate carboxylase/oxygenase) gene from all of the Vitreochlamys strains strongly suggest that V. pinguis from Japan is robustly separated from the clade composed of four strains of V. aulata from the Czech Republic, Slovak Republic and Japan. The sequence data also showed that Vitreochlamys is composed of three clades, which are largely consistent with the ultrastructural features of the stigma and pyrenoids.

The affinity for HCO3− and carbonic anhydrase activity (CA; EC 4.2.1.1) were determined in the red macroalga Bostrychia scorpioides. The affinity for HCO3− was assessed by using three independent methods: the effect of a high pH on photosynthetic rates, photosynthetic conductance for CO2 estimated from the initial slope of photosynthesis rate versus inorganic carbon concentration curves, and the CO2 compensation point. The results obtained from the three methods were consistent in demonstrating that B. scorpioides has a very low affinity for HCO3−, although a certain capacity for its use must not be ruled out. High activities of both external and internal CA were found. Internal CA activity was 10-fold higher than external. External and internal CA were necessary for photosynthesis at pH values higher than 7·0 because O2 evolution was inhibited after adding acetazolamide and ethoxyzolamide (inhibitors of CA). The effect of changing O2 concentration on photosynthesis was also examined. Bostrychia scorpioides exhibited a high sensitivity to O2, demonstrating a C3-like photosynthetic gas exchange physiology. Therefore, the high level of CA activity in B. scorpioides was not enough to support a high affinity for HCO3− and to prevent photorespiration.

The organization of F-actin in vegetative cells of the brown alga Sphacelaria rigidula was studied after staining with a modified rhodamine-phalloidin (Rh-Ph) protocol. The interphase vegetative cells display a well-organized F-actin cytoskeleton, consisting of cortical, endoplasmic and perinuclear arrays of actin filaments (AFs). The organization of these AFs changes slightly during mitosis, while they almost disappear at cytokinesis. The perinuclear AF population becomes more obvious during prophase, especially at the poles. At metaphase, an actin spindle is organized, co-localized with the microtubule spindle, while at anaphase an interzonal AF population appears, which persists at early telophase. At advanced telophase the image changes to a rather diffuse actin meshwork in the mid-area between the daughter nuclei. During post-telophase-early cytokinesis this AF system becomes gradually disassembled and a conspicuous actin plate is formed on the cytokinetic plane. This plate becomes more compact with the progress of cytokinesis, and seems to overcoat the patches of the forming diaphragm, and finally the daughter plasmalemmata. Treatment with cytochalasin B disturbs mitosis, not allowing the cells to proceed to anaphase, and prevents cytokinesis. These observations show that AFs are ubiquitous cytoskeletal elements of the vegetative cells of brown algae. The probable role of AFs during the cell cycle and particularly the unique actin configuration involved in cytokinesis are discussed.

Relationships between phytoplankton growth and supplies of phosphorus and nitrogen have been variously investigated in three lakes in two contrasted English lake areas using physiological nutrient indices (alkaline phosphatase activity, phosphorus debt, surplus phosphorus, phosphorus uptake kinetics, dark ammonium uptake, ammonium uptake kinetics, stimulation of 14C uptake, and batch bioassay) and by inspection of water chemistry. None of the lakes was significantly affected by wastewater effluent but all lay in agricultural catchments. Physiological indices and water chemistry suggested limitation of phytoplankton biomass mostly by phosphorus, and to some extent by nitrogen in summer in North Ormesby Broad, by nitrogen in summer and to some extent by phosphorus in Lily Broad and by nitrogen in White Mere. Indices did not always accord with one another, though were not severely misleading. They added little to conclusions that could be drawn from water chemistry alone. Reasons for the differential relative importance of nitrogen and phosphorus in the two lake areas are discussed. The paradigm of phosphorus limitation, though unchallenged in upland waters and those on poorly weathered rocks, may be less relevant in lowland lakes.

An image analysis system was used to count and measure acetoorcein-stained mitotic chromosomes of the acidothermophilic unicellular red algae Galdieria maxima, G. partita, G. sulphuraria and Cyanidium caldarium (Cyanidiophyceae). Chromosome numbers fell into two groups: n = 2 for all three species of Galdieria and n = 5(7) for C. caldarium. It seems that a separation of C. caldarium from Galdieria is karyologically justified. A chromosome number of 2 appears to be indicative of the genus Galdieria and could be used as a marker to distinguish this taxon from Cyanidium. The two smaller chromosomes in the karyotype of C. caldarium were about 0·4 μm long whereas the other three were 0·5–0·7 μm long. In karyotypes of Galdieria species, the two chromosomes differed in length, the smaller chromosome ranging from 0·8 to 1·8 μm and the larger one from 1·2 to 2·3 μm. The visualization of these extremely small chromosomes was possible due to pretreatment of the cells with the DNA intercalator 9-aminoacridine. The mean absolute length of each chromosome of the three members of Galdieria had statistically significant interspecies differences. Nuclear 1C DNA contents were estimated in the algal cells by the Feulgen technique. All species investigated had genome sizes of 1·50–2·25×10−2 pg. Thus it seems that the members of Cyanidiophyceae have the smallest known genomes of all photosynthetic eukaryotes.

When generating kelp hybrid crosses in vitro, small blades are frequently seen in single-sex cultures of microscopic gametophytes as well as in putative hybrid crosses. We demonstrate a polymerase chain reaction (PCR)-based method that quickly determines the presence/absence of possible parental genomes in the observed blades. ITS1 nrDNA fragments that varied in length in a species-specific way were amplified. Amplification products from possible hybrid blades, apogamic blades and parental gametophytes were compared on agarose gels. Using these methods, we were able to identify a number of apogamic blades (i.e. formed directly from gametophytic cells) as well as a true hybrid sporophyte blade in crosses between Alaria marginata (Alariaceae) and Lessoniopsis littoralis (Lessoniaceae), members of two separate families of the Laminariales. To our knowledge this is the first genotypic confirmation of a hybrid kelp.

The seasonal colonization and vertical zonation of adpressed and upright algae on Phragmites stems in Lake Belau, Germany, was investigated from November 1988 to October 1991. During the autumns and winters from 1988 to 1990 the dominant adpressed species included the diatoms Epithemia adnata, E. sorex, E. turgida and Rhopalodia gibba, while during the same seasons in 1990 and 1991, Cocconeis placentula var. euglypta was abundant. The upright diatoms were most abundant on the middle sections of the stems, whereas those epiphytes with most of their cell surfaces directly in contact with the substratum constituted the greatest part of the biomass on the lowest sections of the stems. Epithemia spp. and R. gibba attained their highest abundances near the sediment as well as buried beneath the most dense periphyton aggregations, while there were dense populations of C. placentula var. euglypta on the entire surface of the stems. Cocconeis is able to tolerate higher light intensities, with photosynthetically active radiation (PAR) values averaging 38 μmol m−2 s−1, than Epithemia and Rhopalodia, which settle at a significantly lower PAR (26 and 20 μmol m−2 s−1, respectively). The change in dominance among the adpressed species from Epithemia and Rhopalodia to Cocconeis can be explained by changes in the grazing habits of the snail Acroloxus lacustris.

The degree of synchrony of light/dark-adapted Chlamydomonas reinhardtii cell-wall-less cells was analysed by flow cytometry. The cultures exhibited the classical growth curve reported for synchronously dividing cells. However, analysis of the nuclear DNA content revealed a partial asynchrony of cell division. This allowed us to determine that cell division lasts only 1 h 15 min. The changes in the distribution of nuclei in G1 and S phases did not exhibit the DNA peaks expected for highly synchronous cells. However, these peaks were observed for the G2+M nuclei and reflected the number of cell divisions. Modification of culture conditions including temperature, photoperiod, quantum irradiance and culture density affected the degree of synchrony but did not completely abolish the asynchrony. An overlap between the divisions of cells dividing one, two or three times could account for the partial asynchrony observed. In fact, it is possible that cells perform their divisions synchronously but that the timing and/or the duration of the cell divisions is different for the three kinds of cells. The synchrony seems to be perturbed when the cells move from the G1 to the S phase. The transduction of the release signal, allowing cells to pass through the restriction point of the G1 phase, might be too slow and insufficiently homogeneous between the cells to allow perfect synchronization. Finally, if highly synchronized cells are needed to study Chlamydomonas reinhardtii cell cycle, other synchronization procedures should be considered.

Time-lapse videomicroscopy was used to observe the effects of various cytoskeletal inhibitors on three important fertilization events in Bostrychia moritziana: spermatial mitosis, gamete fusion (formation of a fertilization pore) and nuclear migration along the trichogyne. The microtubule inhibitor oryzalin disrupted spermatial mitosis but had no other effect on fertilization. The actin inhibitors, jasplakinolide, cytochalasin B, latrunculin A and B and mycalolide B inhibited gamete fusion while BDM, a myosin-disrupting drug, inhibited all three major fertilization events. FL-Phallacidin was used to stain actin filaments in spermatia and trichogynes while microtubules were labelled with antibodies at appropriate stages of fertilization. Microtubules were only evident during spermatial nuclear division. Actin filaments were present in both trichogynes and spermatia throughout fertilization; they formed a discrete ring around the fertilization pore and ensheathed male nuclei as the latter migrated into and along the trichogyne. These results suggest that the actin/myosin system plays a role in the events of fertilization.

The effect of low pH on growth and photosynthesis was compared in two strains of Chlorella isolated from symbiosis with Paramecium bursaria. One, 3N813A, releases large amounts of maltose at low pH; the other, NC64A, does not release maltose in culture. Maltose synthesis at low pH appeared to have a greater effect on cell growth than pH by itself. Growth of maltose-releasing 3N813A showed a direct relationship with pH; growth of non-maltose-releasing NC64A was reduced only as pH fell from 7·0 to 5·5. At pH 5·0, net photosynthetic capacity of 3N813A cells was reduced to 76% of controls at pH 7·0, but the specific growth constant (k) was reduced to 22%, and at pH 4·0 growth and nitrate uptake were inhibited. In 3N813A cells at pH 5·0, carbon (C) was diverted from cell growth, net chlorophyll synthesis, assimilation of nitrogen (N), and synthesis of protein and starch to maltose release, but C[ratio ]N ratios were unchanged. In NC54A cells, starch but not protein synthesis was inhibited at pH 5·0, and a reduction in accumulation of N led to an increase in C[ratio ]N ratios. Unlike N starvation, growth of 3N813A cells at pH 5·0 did not reduce chlorophyll content. When measured at pH 7·0 with excess bicarbonate, photosynthetic parameters of both 3N813A and NC64A cells grown at pH 5·0 were similar to those of cells grown at pH 7·0; however, net photosynthesis of 3N813A cells N starved at pH 7·0 was 22% of that of N-sufficient controls. In symbiosis, release of excess photosynthate as maltose may inhibit cell growth by inhibiting assimilation of inorganic N, but because C[ratio ]N ratios are unchanged, photosynthetic capacity of cells is not affected.

Uncertainties about the identity of type specimens of red algae have frequently led to taxonomic and nomenclatural confusion. A procedure for extracting PCR-amplifiable DNA from formalin-fixed material and herbarium specimens was used to investigate the taxonomic status of several South African Gigartinaceae. We compared nucleotide sequences in the internal transcribed spacer 1 (ITS1) region in type specimens and other historically important collections presently referred to Gigartina bracteata, G. radula and G. stiriata. The following opinions are supported: (1) Fucus bracteatus S.G. Gmelin, Chondrodictyon capense Kützing and Iridaea clathrata Decaisne represent a single species of South African Gigartina (G. bracteata (S.G. Gmelin) Setchell & Gardner) in which the disintegration of tetrasporangial sori results in a reticulate thallus. (2) Mastocarpus polycarpus Kützing, M. incrassatus Kützing and Iridaea lapathifolia Kützing represent a single species of South African Gigartina (G. polycarpa (Kützing) Setchell & Gardner) that has often, but erroneously, been called G. radula (Esper) J. Agardh. (3) Mastocarpus verrucosus Kützing is a later heterotypic synonym of Iridaea papillosa Bory (Sarcothalia papillosa (Bory) Leister) and was based on material that probably came from southern South America rather than from South Africa, the provenance given by Kützing. (4) Fucus stiriatus Turner and Sphaerococcus burmannii C. Agardh represent a single species of South African Sarcothalia (S. stiriata (Turner) Leister).

The cyanobacterium Microcystis PCC7806, grown under continuous light, fermented endogenously stored glycogen to equimolar amounts of acetate and ethanol when incubated anaerobically in the dark. In addition, H2, CO2 and some L-lactate were produced. This fermentation pattern differed from that displayed by cells which had been grown under an alternating light/dark (16/8 h) cycle. Such cells produced much more ethanol than acetate, while no lactate was formed. These differences could not be related to the levels of key enzymes of fermentation, which were identical in the two cultures. The cultures grown under continuous light contained twice as much glycogen as the light/dark-grown cells and the former metabolized it at a rate approximately 3 times as fast as the latter culture. Fermentation in the culture grown under continuous light showed low carbon recovery (59–80%) and high oxidation/reduction balance (approximately 1·5). On the basis of calculations of ATP yield it was concluded that this culture was capable of growth driven by fermentation. The increase in structural cell material would account for the missing carbon and reduction equivalents.

A phylogenetic analysis was conducted on 21 biraphid naviculoid diatom species (from three orders and five families, using 5 protoplast and 21 frustule characters) to test whether the recently described genus Craspedostauros was more closely related to members of the Mastogloiaceae or to other stauros-bearing diatoms. The analysis also tested, and confirmed, the hypothesis that the stauros, as recognized by light microscopy, is a homoplastic character. Whereas partitioned analyses of protoplast and frustule data resolved different parts of the ‘total’ tree, using NONA parsimony analysis of all the data, one tree was obtained. This tree placed Craspedostauros in the mastogloialean clade, rather than near Stauroneis, Staurophora, Haslea or Meuniera. The ordinal and familial groupings suggested by Round and others were also confirmed by the analysis, supported particularly by protoplast and some ultrastructural features.

Phenolic compounds, packaged within vesicles known as physodes, are a major cytoplasmic constituent of eggs and zygotes of the Australasian fucoid Hormosira banksii. Physode movement in early life stages of Hormosira is described. We have used the cytoskeletal inhibitors cytochalasin B, latrunculin B and nocodazole added during different stages of development to investigate the role of the cytoskeleton in movement of these vesicles to regions of active wall formation in the growing rhizoid tip and the plane of cytokinesis prior to cell division. Actin inhibition prevented polarization, cross-wall formation and physode movement. When microtubules were disrupted polarization was still possible, but cross-wall formation and physode movement were prevented. Both actin microfilaments and microtubules are necessary to move physodes to regions of accumulation and active wall formation.

The coldwater dinoflagellate Peridiniella catenata has been examined by light and electron microscopy. The fine-structure resembles that of other dinoflagellates with regard to the dinokaryon, the chloroplast enveloped by three membranes, and the pusule. The flagellar apparatus also resembles that of most other dinoflagellates. The main difference is a missing or reduced striated root connective between the longitudinal microtubular flagellar root and the transverse striated flagellar root. The amphiesma includes a scale-like layer on the outside of the plasmalemma. Another unusual feature is the presence of two distinct size classes of trichocysts, measuring c. 0.2×1 μm and c. 0.7×4 μm, respectively. In their detailed construction they resemble those of other dinoflagellate trichocysts. Peridiniella catenata differs from other gonyaulacoid dinoflagellates in the presence of chloroplasts with single-stalked pyrenoids and in details of the pusular complex. The presence of two size categories of trichocysts and the outer scale-like layer have not been observed in other gonyaulacoid dinoflagellates, indicating that P. catenata is not closely related to these dinoflagellates. However, the presence of two striated collar connectives in P. catenata, a feature hitherto observed only in gonyaulacoids, implies affinity to this group.

A new large DNA virus (PlitV-1) infects the marine filamentous brown alga Pilayella littoralis. It was collected in Alaska and infects other P. littoralis isolates of different geographic origin. The virus has an icosahedral capsid of c. 161 nm in diameter, enclosing an electron-dense core. The genome consists of double-stranded DNA and is approximately 280000 base pairs in size. The virus is latent in somatic cells of the host and is propagated only upon induction of the host's reproductive organs. It causes deformed sporangia, resulting in infertility, and is structurally similar to other brown algal viruses. PCR amplification of a genomic sequence coding for part of a structural glycoprotein of the Ectocarpus siliculosus virus EsV-1 produced a fragment of similar size to that obtained with EsV-1.

A new species of the widespread genus Mesophyllum Lemoine is described. Mesophyllum macedonis nov. sp. grows in shallow (c. 0·5 m depth) waters, tolerating wide temperature fluctuations (12–26 °C) at a salinity of 32–35‰, protected from direct wave and light exposure. The species forms weakly attached rose-coloured clumps with whitish margins on rocky substrata. Its thallus is composed of superimposed foliose lamellae (to 170 μm thick) with dorsiventral organization, non-coaxial hypothallium, limited perithallial growth (to 60 μm thick), rectangular epithallial cells and solitary terminal trichocytes. Tetrasporangial conceptacles are raised and have convex roofs perforated by up to 108 pores that are bordered by filaments composed of slender and occasionally elongated cells. Tetrasporophytes were first observed in 1984 and their occurrence monitored without finding gametophytes or other populations at the type locality or elsewhere on the coast of the Sithonia Peninsula. A comparative study of literature data and material of selected congeners shows that M. macedonis shares most of its characters with M. ornatum (Foslie et Howe) nov. comb. from the Bahamas and Mariana Islands, and M. incisum (Foslie) Adey from southern Australia and New Zealand. Biogeographic scenarios that may explain the restricted distribution of the new Mediterranean endemic are discussed.
