Quantitative real time PCR is the most sensitive and widely used method for the analysis of gene expression. The choice of one or several reference genes is very important for a normalization process, which should not fluctuate under stress conditions, to reduce error rate and bias during experimental procedure. In the present study, the expression stability of nine reference genes (two actins, two tubulins, two elongation factor 1α, two ubiquitins, and cyclophilin) during abiotic stresses such as cold, salt, and PEG treatments, was evaluated on Deschampsia antarctica plants using geNorm software. Results from various experimental conditions indicated that cyclophilin and elongation factor 1α were the most stable genes in the leaf and the root, respectively. The expression of the other reference genes varied under stress. The relative quantification of the TACR7 gene varied according to the kind and the number of reference genes used, suggesting the importance of considering the implications of a combination of reference genes under different stress conditions and in different tissues.

Well-preserved vertebrate microremains are abundant in the residues from a calcareous grey siltstone in the karawaka biozone (?late Givetian) of the Aztec Siltstone at Mount Crean, Lashly Mountains, Antarctica. Acanthodians are represented by scales of acritolepid Pechoralepis juozasi sp. nov., climatiid Nostolepis sp. cf. N. gaujensis Valiukevičius, diplacanthid Milesacanthus antarctica Young & Burrow, and an undetermined acanthodiform. The acanthodian assemblage resembles those from early Frasnian carbonates of central Iran. Chondrichthyan elements in the fauna include rare teeth of Aztecodus harmsenae Long & Young and Antarctilamna prisca Young, ctenacanthoid-type scales and branchial denticles which are possibly from Antarctilamna, and scales of an indeterminate chondrichthyan. An isolated set of acanthodid acanthodiform jaws from the uppermost ‘phyllolepid’ biozone of the Aztec Siltstone at Mount Ritchie, Warren Range, Antarctica is also described.

Two contrasting high nutrient/low chlorophyll regions having different conditions that control phytoplankton production, and separated by an area of blooming, are found during summer in the vicinity of the South Shetland Islands (Antarctica). Low chlorophyll conditions occur either in Fe-rich, deeply mixed and high salinity Weddell Sea shelf waters, or the Fe-poor, shoaled and low salinity Drake Passage Antarctic Circumpolar Current waters, while phytoplankton blooms are located between in mid salinity water. Contrasting phytoplankton communities were found to populate these different biogeochemical provinces. In data from six field seasons (1999–2007), nanoplankton (2–20 μm) were found to be dominant in the phytoplankton populations from light-controlled coastal waters, including blooms, with most chlorophyll found in the 2–5 μm size class. In contrast, the adjacent and presumably Fe-controlled Drake Passage waters were dominated by the microplankton (> 20 μm) size class. The asymmetrical distribution of phytoplankton size classes across the salinity gradient, when analysed independently of total chlorophyll concentration, supports the hypothesis that the different food web grazing dynamics are dependent upon biogeochemical provinces.

After an extended period of sporadic sightings of small numbers of king penguins at the Falkland Islands, they established themselves on Volunteer Point, situated at the north-east of the islands, by the late 1970s. By 1980, a small breeding population was present which yielded some 40 fledglings during that same year. Since 1991, the population has been monitored annually and the resulting fledgling counts analysed to assess population trends. The population demonstrated a significant increase over the past three decades, at about 10% per annum, with time explaining 75% of the variation in count data. The current population is estimated to be 720 breeding pairs. Despite several authors having alluded to the existence of a large colony of king penguins at the Falklands prior to human exploitation, we found no evidence in support of this. We furthermore found no evidence in the literature in support of exploitation for king penguin oil during the 19th century. Unlike at other breeding sites, increasing numbers of king penguins at the Falklands is consequently unlikely to be a recovery response following exploitation, but rather an indication of either increased immigration or of improved feeding conditions.

Ice-free regions of Antarctica are concentrated along the coastal margins but are scarce throughout the continental interior. Environmental changes, including the introduction of non-indigenous species, increasingly threaten these unique habitats. At the same time, the unique biotic communities subsisting in isolation across the continent are difficult to survey due to logistical constraints, sampling challenges and problems related to the identification of small and cryptic taxa. Baseline biodiversity data from remote Antarctic habitats are still missing for many parts of the continent but are critical to the detection of community changes over time, including newly introduced species. Here we review the potential of standardized (non-specialist) sampling in the field (e.g. from soil, vegetation or water) combined with high-throughput sequencing (HTS) of bulk DNA as a possible solution to overcome some of these problems. In particular, HTS metabarcoding approaches benefit from being able to process many samples in parallel, while workflow and data structure can stay highly uniform. Such approaches have quickly gained recognition and we show that HTS metabarcoding surveys are likely to play an important role in continent-wide biomonitoring of all Antarctic terrestrial habitats.

Fossil leaves of the Voltziales, an ancestral group of conifers, rank among the most common plant fossils in the Triassic of Gondwana. Even though the foliage taxon Heidiphyllum has been known for more than 150 years, our knowledge of the reproductive organs of these conifers still remains very incomplete. Seed cones assigned to Telemachus have become increasingly well understood in recent decades, but the pollen cones belonging to these Mesozoic conifers are rare. In this contribution we describe the first compression material of a voltzialean pollen cone from Upper Triassic strata of the Transantarctic Mountains. The cone can be assigned to Switzianthus Anderson & Anderson, a genus that was previously assumed to belong to an enigmatic group of pteridosperms from the Triassic Molteno Formation of South Africa. The similarities of cuticle and pollen morphology, together with co-occurrence at all known localities, indicate that Switzianthus most probably represents the pollen organ of the ubiquitous Heidiphyllum/Telemachus plant.

Within Antarctica there are large gradients both in climate and in vegetation which offer opportunities to investigate links between the two. The activity (% total time active) of lichens and bryophytes in hydric and xeric environments was monitored at Livingston Island (62°39'S). This adds a northern site with a maritime, cloudy climate to previous studies in the southern Antarctic Peninsula and the Dry Valleys (78°S). Annual activity increases northwards from less than 1% to nearly 100%. There is a major and consistent difference between hydric sites which, with snow melt, can be 100% active in summer months even in the Dry Valleys, and xeric sites which, depending on precipitation, rarely exceed 80% activity even at Livingston Island. Mosses dominate hydric sites and lichens the xeric sites all along the gradient. Mean temperatures when active are 2–4°C at all sites, as liquid water is required. Light is a potential major stress reaching 880 µmol m-2 s-1 when active in continental sites. The lack of extremes in temperatures and light combined with high activity levels means that summer at Livingston Island is one of the better sites for lichen and bryophyte growth in the world.

The Antarctic fish fauna from outer continental shelf/upper slope depths is under-sampled compared to that of the inner shelf, and there are limited quantitative data available on absolute abundance and taxonomic change with depth. A photographic survey of demersal fishes was conducted along a depth-gradient of 400–2099 m on the outer shelf and upper slope west of Anvers Island, Palmer Archipelago. A total of 1490 fishes were identified at least to the family level. Notothenioids composed 52.7% of absolute abundance and non-notothenioids 47.3%. The most abundant families were Nototheniidae (39.4%), followed by Macrouridae (28.9%), Zoarcidae (16.9%), and Channichthyidae (12.1%). The most abundant species were the notothenioids Lepidonotothen squamifrons (30.5%) and Chionobathyscus dewitti (11.7%), and the non-notothenioid Macrourus spp. (29.5%). The absolute abundance of all fishes peaked at 400–599 m. Depths of maximum abundance were 400–599 m for L. squamifrons, 700–1499 m for Macrourus spp., and 900–1499 for C. dewitti. At 700–999 m the abundance shifted from primarily notothenioids to the non-notothenioids Macrourus spp. and zoarcids. Fishes of the outer shelf and upper slope are not provincialized like those of the inner shelf and are circum-Antarctic.

Age and diet of 140 brown phase early juvenile Notothenia rossii specimens caught at Potter Cove from December 2005–March 2006 were studied. Fish were immature, in the range 8.5–21 cm TL and of age groups 1–2 (otolith/scale readings). The diet (F% and coefficient Q methods) was mainly benthic-demersal organisms with Gammaridean amphipods (primarily Gondogeneia antarctica) as the most frequent (F% = 98) and main prey (Q% = 97). The occurrence of algae, gastropods and harpacticoid copepods was high, but these organisms were secondary or occasional food. The importance of other benthic (bivalves, polychaetes, isopods) and pelagic (ostracods, calanoids, hyperiid amphipods, krill) prey was very low. Preference of larger fish for larger prey was evident. Stomach fullness analysis showed high food availability during the sampling period. Young stages of N. rossii are demersal, preying mainly on gammarideans and other invertebrates of the benthic community associated with macroalgae beds. The almost complete lack of plankton components in the diet of early juvenile fish suggests they do not migrate vertically to feed.

We examine Weddell Sea deep water mass distributions with respect to the results from three different model runs using the oceanic component of the National Center for Atmospheric Research Community Climate System Model (NCAR-CCSM). One run is inter-annually forced by corrected NCAR/NCEP fluxes, while the other two are forced with the annual cycle obtained from the same climatology. One of the latter runs includes an interactive sea-ice model. Optimum Multiparameter analysis is applied to separate the deep water masses in the Greenwich Meridian section (into the Weddell Sea only) to measure the degree of realism obtained in the simulations. First, we describe the distribution of the simulated deep water masses using observed water type indices. Since the observed indices do not provide an acceptable representation of the Weddell Sea deep water masses as expected, they are specifically adjusted for each simulation. Differences among the water masses’ representations in the three simulations are quantified through their root-mean-square differences. Results point out the need for better representation (and inclusion) of ice-related processes in order to improve the oceanic characteristics and variability of dense Southern Ocean water masses in the outputs of the NCAR-CCSM model, and probably in other ocean and climate models.

A one-year time series of Acoustic Doppler Current Profiler (ADCP) data was collected in Terra Nova Bay (TNB) polynya (Ross Sea, Antarctica) during 2000. Together with Automatic Weather Station (AWS) Eneide meteorological data and Special Sensor Microwave Imager (SSM/I) ice concentration data, ADCP data were analysed to investigate upper layer dynamics and variability due to atmospheric forcing. Empirical Orthogonal Function (EOF) analysis was performed to separate the surface variability caused by local forcing from the large-scale circulation component. In particular, the first mode represented the barotropic circulation while the second the stronger surface currents. The decrease in shelf water density from melting sea ice resulted in an off-shore density gradient producing a southern shift in the circulation. This result proved to be consistent with the in situ data acquired during February–April at 120 m depth. The observed variability of the surface currents was assessed with respect to the thermal wind equation and the steady Ekman model. Strong katabatic winds shifted the surface currents eastward with respect to the general north-eastern circulation. The wind stress acted as a relevant forcing for the surface large-scale circulation in TNB, but had negligible effects on the vertically integrated transport.

Human activity in the Antarctic requires the use of petroleum hydrocarbons as the main energy source for a variety of operations. In the current study, in situ soil microcosms were constructed in the proximity of the Brazilian Antarctic Station Comandante Ferraz, King George Island, South Shetland Islands, to analyse the effect of oil amendment on the indigenous bacterial community in contaminated and uncontaminated sites to assess the potential for bioremediation. Microcosms were sampled for heterotrophic and hydrocarbon-degrader bacterial counts, pH, temperature, moisture, nutrient levels and petroleum hydrocarbons. Total organic carbon, nitrogen and phosphorus contents were generally low in the Antarctic cryosols. According to statistical analysis based on Colony Forming Unit numbers, significant bacterial populations were present in all microcosms, with larger numbers observed in oil amended than in non-amended soils. Aliphatic and aromatic fractions of diesel fuel were detected in the soil microcosms, and significant quantities were removed during the experiment. These results strongly suggest that the cold-adapted bacterial community present in soils around the Brazilian Antarctic station has the potential to adapt and utilize the oil as a carbon source. This knowledge can contribute both to bioremediation technology and the goals of the the Antarctic Treaty which prohibits the introduction of foreign organisms into the region.

The specification, development and deployment of an automated observatory designed to operate unattended for a year in the extreme climatic conditions of the polar regions is presented. Solar- and wind-powered electric generators are used to charge lead/acid batteries to provide a nominal 100 W of electrical power for operating scientific sensors. The equipment is housed in a highly insulated caboose (3m by 2m by 2m). The temperature in the latter is regulated using a unique thermal control system which utilizes the latent heat of 500 l of water stored in 25-l plastic containers. Data are logged to optical disks for collection once per year. The observatory is designed to be air-deployed using a DHC-6 aircraft. The paper discusses the rationale for designing such automated systems, the operational experience gained from several years of operation, and the application of a network of observatories to solar-terrestrial physics research.

In Antarctica, the genus Bartramia has been restricted to a single polymorphic species, B. patens. Its status as a separate species or a subspecies of the Northern Hemisphere B. ithyphylla was debated. In the present paper, we combine analyses of chloroplast (trnS–rps4–trnT–trnL–trnF region) and nuclear ITS sequences with a reinvestigation of morphological characteristics to infer the identity of Antarctic Bartramia. Phylogenetic and Automatic Barcode Gap Discovery (ABGD) species delimitation analyses indicate that the species diversity of Bartramia in Antarctica has been underestimated, since two species were identified, both belonging to Bartramia sect. Pyridium. Of these, B. subsymmetrica is a new record of the species for Antarctica, as it has previously only been recorded from Livingston Island, South Shetlands. The other species is B. patens, which is separated from B. ithyphylla by newly inferred morphological characteristics and is a sister species to the latter in the molecular phylogenetic analyses. Consequently, we consider B. ithyphylla to be a Northern Hemisphere instead of a bipolar species. The suggested conspecificity of both taxa into one species in the ABGD analysis is considered to result from overlumping by this species delimitation method. The delimitation of the three species of section Bartramia (B. halleriana, B. mossmaniana and B. pomiformis) and the circumscription of the genus Bartramia are discussed.

A light, mining drill rig deployed from the stern of a research vessel has been used to carry out shallow drilling in 212 m water depth on the continental shelf in the eastern Weddell Sea. Penetration was 15 m below the seabed with 18% recovery in the 31 hours available for the experiment. The recovered glacigenic sediments are predominantly volcanic material of basaltic and andesitic composition with petrological characteristics and age similar to the continental flood basalts exposed in Vestfjella, about 130 km upstream from the drill site. The sediments include a reworked marine Miocene diatom flora. The material documents oscillations of the East Antarctic Ice Sheet over the past 30 ka. The lowermost diamicton probably represents a deformation till, and the grounding line retreated past the drill site 30 km from the shelf edge about 30 kyr BP. A readvance occurred during the Late Wisconsin Glacial Maximum. Assuming a reservoir correction of 1300 yr, marine conditions existed at the site between 10.1-7 kyr BP, and later at least between 2.8 and 2.5 kyr BP. The stratigraphy at the site has been disturbed by iceberg ploughing and/or contact between the ice shelf and the sea floor during local advances after 2.5 kyr BP.

Values of attenuation coefficient, Kd(λ), for five visible wavelengths are reported for 14 sites around South Georgia and the Bransfield Strait, in the Southern Ocean. The mean chlorophyll-plus-phaeopigment concentrations in the upper 30 m of the water column ranged from 0.32–6.633 mg m−3 with one particularly high mean value of 31 mg m−3. Partition of attenuation between chlorophyll (Kc) and other factors in the water column (Ko) indicated that the spectral character of Ko in the Bransfield Strait was consistent with absorption by non chlorophyll-like pigments (Gelbstoff). Values were significantly different between the two areas. Values of the specific attenuation coefficient due to pigment concentration (Kc) were small compared to temperate values. These data support other studies in suggesting that the application of remotely sensed ocean colour data to global biogeochemical surveys requires the development of regional algorithms.

We have investigated how the microbially-driven processes of carbon (C) mineralization (respiration) and nitrogen (N) mineralization/immobilization in a soil from the northern Maritime Antarctic respond to differences in water availability (20% and 80% water-holding capacity) and temperature (5°C and 15°C) in the presence and absence of different organic substrates (2 mg C as either glucose, glycine or tryptone soy broth (TSB) powder (a complex microbial growth medium)) in a controlled laboratory experiment over 175 days. Soil respiration and N mineralization/immobilization in the presence of a C-rich substrate (glucose) increased with increases in water and temperature. These factors were influential individually and had an additive effect when applied together. For the N-rich substrates (glycine and TSB), microbial responses to increased water or temperature alone were weak or not significant, but these factors interacted to give significantly positive increases when applied together. These data indicate that under the expected changes in environmental conditions in the Maritime Antarctic, where temperature and the availability of water and organic substrates will probably increase, soil microbial activity will lead to more rapid C and N cycling and have a positive feedback on these biogeochemical processes, particularly where or when these factors increase concurrently.

Rates of tag-loss are determined for Dalton Jumbo Rototags applied to the hind flippers of 4343 (2208 males, 2135 females) southern elephant seal (Mirounga leonina) pups at Marion Island over an eight year period from 1983–1990 as part of a demographic study of the species. Loss rates were the lowest recorded to date for this species (range 0.0–9.1%). No significant relationship existed between age and rate of tag-loss, neither was there any sex or year related differences in age-specific tag-loss rates. The low rates of loss highlight the value of tagging as a marking technique, and allow for high levels of confidence in the reliability of the population parameters that are derived from the tagging data collected for the Marion Island population.

Measurement of pressure fluctuations provides the best means of detecting atmospheric internal gravity waves at the Earth's surface. We have developed an instrument which is sufficiently sensitive to detect the small pressure fluctuations associated with such waves yet robust enough for deployment in an Antarctic environment. The instrument incorporates several novel features, both in its design and in the method of deployment used. A prototype instrument has been successfully deployed at the British Antarctic Survey's Halley Station during 1989. The design of an experiment using an array of six improved instruments is briefly described.
