A growing body of literature has explored the influence of physical activity on brain structure and function. While the mechanisms of this relationship remain largely speculative, recent research suggests that one of the effects of physical exercise is an increase in synaptic long-term potentiation (LTP). This has not yet been explored directly in humans due to the difficulty of measuring LTP non-invasively. However, we have previously established that LTP-like changes in visual-evoked potentials (VEPs) can be measured in humans. Here, we investigated whether physical fitness status affects the degree of visual sensory LTP. Using a self-report measure of physical activity, participants were split into two groups: a high-activity group, and a low-activity group. LTP was measured and compared between the two groups using the previously established electroencephalography-LTP paradigm, which assesses the degree to which the N1b component of the VEP elicited by a sine grating is potentiated (enhanced) following a rapid “tetanic” presentation of that grating. Both groups demonstrated increased negativity in the amplitude of the N1b component of the VEP immediately after presentation of the visual “tetanus,” indicating potentiation. However, after a 30-min rest period, the N1b for the high-activity group remained potentiated while the N1b for the low-activity group returned to baseline. This study presents the first evidence for the impact of self-reported levels of physical activity on LTP in humans, and sheds light on potential neurological mechanisms underlying the relationship between physical fitness and cognition. The aim was to investigate the temporal response properties of magnocellular, parvocellular, and koniocellular visual pathways using increment/decrement changes in contrast to elicit visual evoked potentials (VEPs). Static achromatic and isoluminant chromatic gratings were generated on a monitor. Chromatic gratings were modulated along red/green (R/G) or subject-specific tritanopic confusion axes, established using a minimum distinct border criterion. Isoluminance was determined using minimum flicker photometry. Achromatic and chromatic VEPs were recorded to contrast increments and decrements of 0.1 or 0.2 superimposed on the static gratings (masking contrast 0–0.6). Achromatic increment/decrement changes in contrast evoked a percept of apparent motion when the spatial frequency was low; VEPs to such stimuli were positive in polarity and largely unaffected by high levels of static contrast, consistent with transient response mechanisms. VEPs to finer achromatic gratings showed marked attenuation as static contrast was increased. Chromatic VEPs to R/G or tritan chromatic contrast increments were of negative polarity and showed progressive attenuation as static contrast was increased, in keeping with increasing desensitization of the sustained responses of the color-opponent visual pathways. Chromatic contrast decrement VEPs were of positive polarity and less sensitive to pattern adaptation. The relative contribution of sustained/transient mechanisms to achromatic processing is spatial frequency dependent. Chromatic contrast increment VEPs reflect the sustained temporal response properties of parvocellular and koniocellular pathways. Cortical VEPs can provide an objective measure of pattern adaptation and can be used to probe the temporal response characteristics of different visual pathways.

Previous studies report that background luminance flicker, which is asynchronous with signal averaging, reduces the amplitude and increases the latency of the pattern-onset visual evoked potential (VEP). This effect has been attributed to saturation of the magnocellular (m-) pathway by the flicker stimulus. In the current study, we evaluate this hypothesis and further characterize this effect. We found that flicker had similar effects on the pattern-onset and pattern-reversal VEP, suggesting that the reversal and onset responses have similar generators. Chromatic flicker decreased latency of the chromatic VEP whereas luminance flicker increased peak latency to luminance targets. This result indicates that luminance flicker saturates a rapidly conducting m-pathway whereas chromatic flicker saturates a more slowly conducting parvocellular (p-) pathway. Finally, evoked potentials to chromatic and luminance stimuli were recorded from 34 electrodes over the scalp in the presence of static and asynchronously modulated backgrounds. An equivalent dipole model was used to assess occipital, parietal, and temporal lobe components of the surface response topography. Results showed that chromatic flicker reduced activity to a greater extent in the ventral visual pathway whereas luminance flicker reduced activity to a greater extent in the dorsal visual pathway to parietal lobe. We conclude that the VEP to isoluminant color and luminance stimuli contains both m- and p-pathway components. Asynchronous flicker can be used to selectively reduce the contribution of these pathways to the surface recorded VEP. Our results provide evidence of parallel pathways in the human visual system, with a dorsal luminance channel projecting predominantly to the posterior parietal lobe and a ventral color channel projecting predominantly to inferior temporal lobe.

Positive- and negative-contrast stimuli yield the perceptions of brightness and darkness, respectively, and are processed separately by ON and OFF neural pathways. The properties of these morphologically and pharmacologically distinct subsystems were measured in humans by recording visual evoked potentials (VEPs). These electrical responses from the visual cortex were elicited by novel positive- and negative-contrast stimuli, designed to emphasize, selectively, contributions from ON and OFF pathways. Results revealed differential processing of the two types of contrast information, suggesting asymmetries in ON and OFF subsystems; OFF subsystems have finer spatial tuning and greater contrast gain than ON subsystems. These VEPs may be useful in diagnosing neurological disorders that involve primarily one subsystem.

We measured transient and steady-state checkerboard contrast-reversal visual evoked potentials (VEPs) in ten dyslexics, five patient controls, and 11 normals over a range of contrasts and luminances. Latency, amplitude, and phase measurements failed to distinguish the responses of dyslexics from those of normals or patient controls. Decreases in luminance or contrast resulted in an increased latency of the transient VEP in all groups, but these changes also did not distinguish the responses of dyslexics from those of the controls. Response variability was similar in dyslexics and normals, but was increased in subjects with attention deficit-hyperactivity disorder (ADHD). Performance on standardized psychometric testing did differentiate the dyslexics from controls, but did not correlate with VEP responses.

The purpose of the present study was to compare standard multifocal visual evoked potential (mfVEP) pattern-reversal responses with those produced by pattern-onset, pattern-offset, and pulsed pattern stimuli. mfVEP recordings were obtained from five normal subjects using VERIS and a 4-electrode array. The standard reversal stimulus had 215 m-sequence steps (7.5-min duration). Pattern-onset and -offset responses were evaluated using sequences that all had 32 frames per m-step and 210 total steps (7.5 min); but the duration of the contrast step varied so that it was 1, 2, 4, 8, 12, or 16 of the 32 frames. The same series was also inverted so that adapting contrast was high and the stimulus step began with a contrast decrement. The effect of temporal sparseness was studied with positive contrast pulses (two-frame duration) within 16, 20, 24, 28, or 32 frames per m-step (all had 211 total steps). Four stimulus locations were isolated to study the effect of spatial sparseness. Standard mfVEP reversal responses were virtually identical to onset responses throughout the field, but ~3.5 times smaller. Responses to pattern onset were about twice as large as those for offset, especially in the lower hemifield, irrespective of adapting contrast level. Offset responses exhibited a different waveform compared with reversal, onset, or brief pulse responses. Though temporally sparse pattern-pulse responses were ~3.5 times larger than standard reversal responses, there was no improvement in signal-to-noise ratio (SNR). However, spatial isolation increased SNR by 22% for reversal responses and by 62% for temporally sparse pulses. Temporally sparse pattern-pulse stimuli do not improve mfVEP SNR unless they are also spatially sparse, suggesting that lateral inhibitory mechanisms have a greater impact than temporal contrast gain control mechanisms. The mfVEP response depends on the polarity of a contrast step, irrespective of the state of contrast adaptation.

The purpose of this study was to specify isoluminance at different retinal eccentricities and to characterize macular pigment optical density (MPOD) and distribution using the steady-state visual evoked potential (VEP). Red–green (R/G) and blue–green (B/G) gratings were generated within two circular stimulus fields (radius = 0.55 or 1.1 deg) and within four annular fields (maximum mean radius = 6.0 deg) on a color monitor. Temporal frequency was 15 Hz. Isoluminance was determined for each stimulus using minimum flicker photometry. Steady-state onset–offset VEPs were recorded to the same annular stimuli as the luminance ratio between adjacent chromatic components was changed from 0.25 to 0.85 in 11 automated steps (0.5 representing photometric isoluminance). Fourier analysis showed that the power of the first harmonic was minimized at the isoluminant ratio specific to each subject. Relative OD was computed by comparing the isoluminant ratio at any location with that for the most eccentric annulus. To compensate for the broadband characteristics of the monitor, OD values were corrected according to minimum flicker measurements made through known concentrations of carotenoid solution. MPOD was additionally measured using minimum motion photometry.

There was high correlation between the isoluminant ratios determined by minimum flicker and VEPs for both R/G and B/G stimulation (r = 0.91, P < 0.005, slope = 1). Calibrated OD values computed from VEP estimates of B/G isoluminance correlated with those derived from minimum flicker (r = 0.96, P < 0.0005, slope = 0.85) and motion photometry (r = 0.94, P < 0.0005, slope = 0.88). OD values derived from B/G VEPs increased toward the fovea and corresponded closely with minimum flicker and minimum motion assessment of MP distribution profiles. The steady-state VEP can be used to determine isoluminance at different retinal eccentricities. MPOD and distribution can be measured by steady-state VEPs to B/G stimuli.

It would be informative to have an electrophysiological method to study, in an objective way, the effects of mercury exposure and other neurotoxics on human color vision performance. The purpose of the present work was to study human color discrimination by measuring chromatic difference thresholds with visual evoked potential (VEP). Six young normal trichromats (24 ± 1 years old) and one deutan (26 years old) were tested. The stimuli consisted of sinusoidal isoluminant chromatic gratings made from chromaticity pairs located along four different color directions centered on two reference points. Heterochromatic flicker photometry (HFP) protocol was used to obtain the isoluminance condition for every subject and for all chromaticity pairs. Spatial frequency was 2 cycles/deg. Presentation mode comprised onset (300 ms)/offset (700 ms) periods. As previously described, we found a negative deflection in the VEP which was related to the chromatic difference: as chromatic difference increased, amplitude increased and latency decreased. VEP response amplitude was plotted against distance in the CIE 1976 color space between the grating chromaticities and fitted with a regression line. We found color thresholds by extrapolating the fitting to null amplitude values. The thresholds were plotted in the CIE 1976 color space as MacAdam ellipses. In normal trichromats the ellipses had small size, low ellipticity, and were vertically oriented. In the deutan subject, the ellipses had large size, high ellipticity, and were oriented towards the deutan copunctal locus. The VEP thresholds were similar to those obtained using grating stimuli and psychophysical procedures, however smaller than those obtained using pseudoisochromatic stimuli (Mollon-Reffin method). We concluded that transient VEP amplitude as a function of contrast can be reliably used in objective studies of chromatic discrimination performance in normal and altered human subjects.

Previous research in adults has demonstrated the utility of the visual evoked potential (VEP) to measure the integrity of the chromatic and achromatic visual pathways. The VEP has also been shown to be a valuable indicator of maturation of these pathways in infants up to 1 year of age. The present manuscript reports changes in the visual pathways from 2 years to adulthood as measured by the spatio-chromatic VEP. The responses to achromatic reversal stimuli designed to preferentially activate the low spatial-frequency achromatic (luminance) pathways appear adult-like by 1 year of age. The responses to low spatial-frequency isoluminant onset stimuli designed to preferentially activate the chromatic pathway do not appear as they do in the adult until after 12–13 years of age. The shapes of the chromatic VEP waveforms shift from a positive–negative complex to a negative–positive complex. These changes can be modeled by a decrease in the latency of a large negative component between the ages of 1 year and adulthood. The results suggest that for low spatial-frequency stimuli, there are long-term changes in the development of the chromatic pathways that are not observed in the low spatial-frequency achromatic pathways. The changes in the chromatic VEP waveforms with age may be a physiological correlate of reported behavioral changes.

Mechanisms of motion vision in the human have been studied extensively by psychophysical methods but less frequently by electrophysiological techniques. It is the purpose of the present investigation to study electrical potentials of the eye (electroretinogram, ERG) and of the brain (visual evoked potential, VEP) in response to moving regular square-wave stripe patterns spanning a wide range of contrasts, spatial frequencies, and speeds. The results show that ERG amplitudes increase linearly with contrast while VEPs, in agreement with the literature, show an amplitude saturation at low contrast. Furthermore, retinal responses oscillate with the fundamental temporal stimulus frequency of the moving pattern while brain responses do not. In both the retina and the brain, the response amplitudes are tuned to certain speeds which is in agreement with the nonlinear correlation-type motion detector. Along the ascending slopes (which means increasing amplitudes) of the tuning functions, the ERG curves overlap at all spatial frequencies if plotted as a function of temporal stimulation frequency. The ascending slopes of the tuning functions of the VEP overlap if plotted as a function of speed. The descending slopes (which means decreasing amplitudes) of the tuning functions show little (ERG) or no (VEP) overlap and the waveforms at high speeds approach pattern-offset-onset responses. These observations suggest that in the retina motion processing along the ascending slopes of the tuning curves takes place by coding the temporal stimulation frequency which depends on the spatial frequency of the moving pattern. In the brain, however, motion processing is by speed independent of spatial frequency. Simple calculations show that the VEP information is decoded from the ERG signal into a speed signal.

This study used the swept spatial-frequency method to compare retinal and cortical acuity in anesthetized young adult rhesus monkeys. Visual evoked potentials (VEPs) and pattern electroretinographic responses (PERGs) were recorded from 25 monkeys (age range: 4–12 years) anesthetized with a continuous infusion of propofol. The stimuli were temporally countermodulated sine-wave gratings that increased in spatial frequency within a 10.24-s period. All animals were refracted using acuity estimated from the zero micro-volt intercept of the linear regression of evoked potential amplitude on spatial frequency. Average sweep acuities were 23.7 cycles/deg ± 1.5 S.E.M. and 23.1 cycles/deg ± 1.8 S.E.M. for the PERG and VEP, respectively. VEP and PERG acuities were within the range expected based on acuities estimated from behavioral studies in macaques. PERG and VEP acuities were highly correlated (r = 0.90) and equally sensitive to spherical blur. On a subset of animals, test–retest reliability of animals, and interocular correlations, were high (r = 0.87 and r = 0.83, respectively). Increasing propofol dosage 8-fold did not degrade PERG or VEP acuity. This study demonstrates that high spatial-frequency acuities can be rapidly obtained from young adult rhesus monkeys under a wide dose range of propofol anesthesia using the swept spatial-frequency method.
